À l'issue d'une mutagenèse, on dispose d'une collection d'allèles mutants. En général, ces allèles confèrent le même phénotype. La question se pose alors de savoir combien de gènes sont mis en jeu pour obtenir cette collection d'allèles. La génétique a développé des méthodes qui permettent de répondre à cette question. Il existe deux méthodes : la complémentation et la cartographie. Nous allons commencer dans ce chapitre par le test de complémentation. C'est en effet le plus rapide à mettre en œuvre et son application permet d'avoir une bonne idée du nombre de gènes. Le test consiste dans la fabrication d'une souche pour laquelle les allèles mutants sont associés deux à deux en configuration trans et en l'observation du phénotype. Dans le cas de mutants biochimiques, par exemple, ce phénomène se traduit par le fait que l'hétérozygote pousse faiblement sur milieu minimum. L'explication la plus fréquemment valable est que la molécule enzymatique, dont le cistron concerné est le gène de structure, est un oligomère formé de plusieurs polypeptides identiques. When the mutations occur on the same chromosome(cis position) the phenotype is wild type; when the mutations are on different chromosomes(trans position) the mutant phenotype emerges. L'effet de position cis-trans le plus courant correspond au cas où les deux sites mutés appartiennent au même cistron. Lorsqu'ils sont en position cis, l'un des deux cistrons homologues n'a subi aucune mutation et le phénotype est normal.
