The procedure is based on the principle that two digital radiographic images obtained under different time intervals, with the same projection geometry, are spatially and densitometrically aligned using specific software. When the two images are registered and intensities of corresponding pixels are subtracted of the gray scale values, a uniform difference image is produced, resulting in a new image representing the differences between the two, called the subtraction image. SIAM is a technology that defies the current laws of imaging. It permits scientists to view images much smaller than the wavelength of light and approaching the atomic level, an achievement that has long been considered impossible according to the basic principles of classical optics. SIAM works by shining light through a thin sample onto an ultra-sharp, vibrating tip on the other side. The light scattering from the tip gets altered as the tip approaches within a nanometer from the sample surface, as it vibrates, due to the optical interaction with the local properties of the sample. An image is built up by recording the scattered light as the probe is scanned line by line. The scientists recorded the variations in the scattered light using a very sensitive device--an optical interferometer--which is capable of measuring modulations in light level corresponding to single photons. Ces forces modifient les vibrations de la lamelle. Après s'être réfléchi sur l'échantillon et sur la lamelle, le faisceau d'analyse se combine avec le faisceau de référence dans un dispositif interférométrique. Un nouveau faisceau permet d'analyser les modifications des vibrations de la lamelle; il renseigne donc sur le relief de l'échantillon observé. Scanning transmission electron microscopy(STEM) combines the principles of transmission electron microscopy and scanning electron microscopy and can be performed on either type of instrument. Like TEM [transmission electron microscopy], STEM requires very thin samples and looks primarily at beam electrons transmitted by the sample. One of its principal advantages over TEM is in enabling the use of other signals that cannot be spatially correlated in TEM, including secondary electrons, scattered beam electrons, characteristic X-rays, and electron energy loss. Like SEM [scanning electron microscopy], the STEM technique scans a very finely focused beam of electrons across the sample in a raster pattern. Interactions between the beam electrons and sample atoms generate a serial signal stream, which is correlated with beam position to build a virtual image in which the signal level at any location in the sample is represented by the gray level at the corresponding location in the image.
