ß-Sheets are formed when two or more almost fully extended polypeptide chains are brought together side by side, so that regular hydrogen bonds can form between the peptide backbone amide NH and carbonyl oxygen groups of adjacent chains.... All the peptide linkages participate in this cross-linking and thus lend the structure great stability; the R groups lie above or below the zigzagging planes of the pleated sheet.... Immobilization of enzymes by crystallization and subsequent cross-linking provides structures which have a regular three-dimensional molecular arrangement, and a high packing density. Such structures are useful for both X-ray diffraction analysis, and also physico-chemical studies of enzymes in the solid state. An important feature of the enzyme crystals is that they have large liquid-filled channels surrounding the individual protein molecules, therefore, as in dilute solution, most of the surface of the enzyme is in contact with the surrounding solvent. Since the channels are permeable to low molecular weight solutes, it is possible to study the properties of enzymes in crystals in much the same way as when in solution. Lysosyme is an... enzyme whose catalytic mechanism is being studied. X-ray analysis of the crystalline enzyme shows that it contains a long cleft as its active site, which can accommodate its normal substrate, the long-chain peptidoglycan of bacterial cell walls. 3, record 12, English, - crystalline%20enzyme

Under the action of thrombin, which splits off a small peptide from fibrinogen, fibrinogen is activated and starts to interact with other monomers. Analysis of the surface of the crystallographic models of most proteins should target at least one surface residue whose replacement would substantially increase the stability of the protein.... Unfortunately, most enzymes do not presently have a crystallographic model available for identification of one or more residues having exposed apolar side chains.... For investigators having neither a crystallographic model of the enzyme of interests or a facility for obtaining and engineering the structural gene, use of a cross-linking reagent is recommended to increase stability. This approach is based on diminishing the polypeptide entropy that is the principal thermodynamic quantity stabilizing the denatured form. 2, record 21, English, - crystallographic%20model

Methods for enzyme immobilization can be classified into three basic categories as follows. 1) Carrier-binding method : the binding of enzymes to water-insoluble carriers. 2) Cross-linking method : intermolecular cross-linking of enzymes by means of bifunctional or multifunctional reagents. 3) Entrapping method : incorporating enzymes into the lattice of a semipermeable gel or enclosing the enzymes in a semipermeable polymer membrane. 2, record 22, English, - enzyme%20immobilization

The technique has two advantages: expensive enzymes can be recovered and used again; the enzyme can be used in a variety of configurations of bioreactors that permit continuous operation. These include packed bed continuous-fed columns and expanded bed systems. A wide range of techniques are used for enzyme immobilization. These are similar in many ways to the techniques used to produce preparations of immobilized cells. 1, record 22, English, - enzyme%20immobilization

A urethane is formed from the reaction of an isocyanate with an alcohol. In general, polyurethanes are made using a diisocyanate, a low molecular weight diol(thermoplastic) and/or triol(cross-linked), and a polyether glycol or a polyester glycol. If the reagents are difunctional, a non cross-linked thermoplastic polyurethane will be formed; if the functionality of at least one of the reagents is greater than two, a cross-linked or three dimensional network will form. Cross-linked polyurethanes exhibit less mechanical creep than non cross-linked polyurethanes. The rubbery modulus of polyurethanes increases as the level of cross-linking is increased. Polyurethanes are diblock copolymers. They consist of hard segments(isocyanate and the chain extender rich areas) that act as physical crosslinks, and soft segments(polyether or polyester glycol rich areas). 3, record 28, English, - three%20dimensional%20network

Thermoset molding. Thermosetting materials can be molded on reciprocating screw machines. The screw is designed differently and the barrel is usually heated with hot water. When thermosets are molded, the heating cylinder provides just enough heat to start exothermic cross-linking chemical reactions between the two monomers. The material must be transferred quickly into the mold and cannot be permitted to cool in the cylinder. If it does, the screw has to be removed and the thermosetting material chipped out. 1, record 46, English, - thermoset%20molding

Trichohyalin used in conjunction with transglutiminase forms a naturally-occurring proteinaceous gel with potential application in the areas of food production/stabilization, cosmetics and coverage for open wounds and burns. We have demonstrated that, using cloned cDNAs, the combination of human trichohyalin with an enzyme that is capable of cross-linking proteins can produce a stable, quickly-formed proteinaceous gel. This technology may be useful for the treatment of skin diseases may have benefit as a transglutaminase replacement therapy. [Source Internet]. 1, record 48, English, - transglutaminase

Cross-linking of IgE receptors(under physiological conditions by antigen, under experimental conditions by antibodies to IgE, by antibodies to receptor, or by IgE aggregates) is required for receptor redistribution and for internalization of receptor. When receptor-bound IgE molecules bind to multivalent antigen they become indirectly cross-linked, they initiate histamine release from the cell, and the IgE molecules together with their receptors and their bound antigen are internalized... A number of biochemical events have been studied that occur immediately after the apposition(or "cross-linking") of adjacent IgE receptors. 1, record 50, English, - cross%2Dlinking

1, record 55, French, - structure%20d%27une%20enzyme
