Method devised by Brown which is particularly useful when, owing to its chemical nature, insolubilization of an enzyme cannot be achieved by glutaraldehyde crosslinking alone. To eliminate the loss of enzyme activity due to diffusion effects caused by crosslinking, reactions enzymes, at low concentrations, are crosslinked via glutaraldehyde to a nonenzymic protein which is rich in L-lysine residues(such as bovine serum albumin), and the resultant immobilized enzyme can be insolubilized by gel entrapment methods.

7, record 8, French, - caoutchouc%20thermoplastique

There has been interest in applying nanogels to drug delivery systems, such as protein delivery and gene delivery. In general, chemically cross-linked nanogels are synthesized by microemulsion polymerization or a crosslinking reaction of intra-associated polymer molecules. We develop tailor-made functional nanogels to create novel nanobiomaterials(nanogel engineering) by the self-assembly of functional associating polymers as building blocks. In particular, nanogels of cholesteryl group-bearing pullulan(CHP) selectively interact with proteins as a host and are useful as artificial molecular chaperone and drug carriers such as cancer immune therapy. 3, record 14, English, - nanogel

Crosslinking... involves the addition of the appropriate amount of crosslinking agent to an enzyme solution under conditions which give rise to the formation of multiple covalent bonds.... The reagents required for crosslinking possess two identical functional groups(homobifunctional reagents) or two or more different functional groups(heterobi-or heteromulti-functional reagents), the latter being more common in binding enzymes to insoluble carriers than in intermolecular crosslinking reactions. The main reagents used contain : carboxyl functional groups,... diazo groups,... isocyanate groups,... alkyl iodides and iodoacetamides. 1, record 15, English, - crosslinking%20agent

The most commonly employed method for preparing water-insoluble enzyme derivatives is through intermolecular crosslinking of enzyme molecules with multifunctional reagents.... This immobilization method is based on the formation of chemical bonds, as in the covalent binding method, but water-insoluble carriers are not used in this method.... The correct choice of reaction conditions is indeed one of the major considerations in dealing with the chemical modification of proteins with multifunctional reagents for producing water-insoluble enzyme derivatives. The concentration of the enzyme and the reaent, the pH and ionic strength of the solution, etc. must be chosen carefully so as to be favorable for intermolecular crosslinking between enzyme molecules. 2, record 19, English, - intermolecular%20crosslinking

In many cases the presence of an insoluble or even soluble carrier matrix may become an insuperable obstacle for the enzyme to perform its function in the organism. In such cases investigators have used bifunctional reagents to stabilize enzymes by intramolecular crosslinking. As a rule these attempts have had only limited success, based on trials with many enzymes.... The matter is complicated by the fact that the bifunctional reagent can interact with the enzyme in three different ways : one point modification, intermolecular linking, and intramolecular crosslinking. 1, record 30, French, - inclusion%20dans%20un%20gel

The most suitable gel material requires both a relatively high concentration of the monomer(acrylamide) to give mechanical rigidity and a high concentration of crosslinking reagent(N, N. methylenebisacrylamide) to achieve the highest possible yield of immobilized enzyme activity.... The trapping efficiency of polyacrylamide gels of varying composition was examined by several investigators. Degani and Miron observed that the maximum yield of activity trapping(56%) occurred with cholinesterase at a crosslinking concentration of 5% and with a total and constant monomer concentration of 15%. 1, record 31, English, - activity%20trapping

Peresters.
