Type of chromatography developed following the observation that certain proteins were unexpectedly retained on affinity gels containing hydrophobic spacer arms. This concept was extended, and families of hydrophobic adsorbents prepared using a homologous series of hydrocarbon chains of different lengths. In practice most proteins can be satisfactorily purified using C-8 or phenyl substituents. Hydrophobic interactions are strongest at high ionic strength, so adsorption to a hydrophobic matrix can be conveniently performed after a salt precipitation or ion exchange chromatography step. Large amounts of unsold bread, doughnuts, cakes and other pastries are available in some areas and are excellent energy sources for ruminant rations. They are usually high in fat and low in crude fibre. Protein levels(on a dry-matter basis) in the range of 10-12% are typical. The low fibre content of the baked material and the baking process itself result in a feed which tends to stimulate ruminal propionate and reduce ruminal acetate production. Scrapie, the prototype of a group of diseases which have the unique property of being both hereditary and infectious, is also exceptional in that it fails to evoke an immune response. Purification of crude scrapie preparations showed a strong association of infectivity with a membrane protein(’PrPSC') ;but a protein with the same amino acid sequence(’PrPC') was subsequently also found in normal mammalian nervous tissue. It is postulated by some investigators that ’PrPSC' is itself the infectious agent, or the most important part thereof, but in papers making that proposal immunological aspects have not been addressed. Experimental evidence supporting the hypothesis of a membrane fragment as agent has likewise lately not been taken into account.
