Short tandem repeat polymorphisms, also referred to as variable number of tandem repeats(VNTRs), are very important to scientists, because they can be used to identify individuals genetically. Marker systems are used to detect loci that respond to restriction enzyme cleavage by producing a fragment that can have many different lengths. This polymorphism is due to variation in the number of tandem repeats. VNTRs can be identified in various ways. Probes can be developed which will hybridize to complementary sequences after RFLP has been performed on a sample of genomic DNA(Southern blots). A second method involves the use of PCR. If the sequence surrounding the VNTR is known, PCR primers(oligonucleotides) can be designed to amplify the repeat region(s). [Plasmids] generally account for only a small fraction of the total host cell DNA, they often carry vital genes, such as those that confer resistance to antibiotics. Because it is so much smaller, plasmid DNA can easily be separated from the DNA of the host cell and purified. For use as cloning vectors, such purified plasmid DNA molecules are cut once with a restriction nuclease and then annealed to the DNA fragment that is to be cloned. The hybrid plasmid DNA molecules produced are then reintroduced into bacteria that have been made transiently permeable to macromolecules. Only some of the treated cells will take up a plasmid. They can be selected by the antibiotic resistance conferred on them by the plasmid since they alone will grow in the presence of antibiotic. Fluorescence-based amplified fragment length polymorphism(fbAFLP) is a novel assay based on the fluorescent analysis of an amplified subset of restriction fragments. The fbAFLP assay involves the selective PCR [polymerase chain reaction] amplification of restriction fragments from a total digest of genomic DNA. The ligation of adapters with primer-specific sites coupled with primers containing selective nucleotides allowed the full potential of PCR to be realized while maintaining the advantages of restriction endonuclease analysis. Avipoxviruses, including Fowlpoxvirus (FPV) have been shown previously to infect mammalian cells but are unable to complete their replication cycle. Avipoxvirus based rabies and HIV vaccines have shown promise in preclinical studies. We have begun work to develop the FPV system for application to HPV CTL. Primed in situ labeling(PRINS) is a fast alternative to traditional in situ hybridization, requiring only a tew handling steps(1, 2).
