In photoabsorption spectroscopy, the intensity of monochromatic light passing through a sample is measured, as a function of wavelength(or energy), using a photosensitive detector like a photomultiplier or a photodiode. A decrease in this transmitted intensity, which appears as a dip in the baseline of incident intensity, when intensity is plotted versus wavelength, indicates the occurrence of an electronic or molecular transition in the sample.

CONT

Confocal microscopy. Unlike "normal" microscopy, only a spot of a minimum size in the specimen is illuminated in confocal microscopy. To enable an image to be formed from this, the specimen must be scanned. The image of the spot is directed through a pinhole stop in an intermediate image plane. As a result, only light from the focal plane can reach the detector(a photomultiplier). All other(out-of-focus) planes are blocked out. This results in an "optical section". The images are stored electronically and displayed on a monitor. A series of optical sections can be recorded by moving a motor a slight distance along the z-axis each time an image has been recorded, after which the next image is then also recorded. Such a z-series permits the electronic reconstruction of the three-dimensional structure using suitable computer programs. The procedure, which is restricted to incident-light techniques, has completely revolutionized fluorescence microscopy in biology in particular.

CONT

... a spectroscope or spectrograph... are effectively the same thing except that a spectroscope is simply used for visual inspection(that is, your eye is the detector), while a spectrograph includes some means(photographic film or an electronic detector) for recording the spectrum for analysis. In professional astronomy these days there is very little need for a spectroscope... a spectrograph in its simplest form,... is a light-tight box with a small(often narrow rectangular or adjustable) opening to let light in, a grating to break the light into its components, and a "detector" of some kind placed at the proper angle and distance from the grating to record the spectrum of the wavelength range of interest. Telescopes are used to gather the faint light from distant objects, and the spectrographs are placed at the focus of the telescope to analyze the light.

... any unwanted disturbance in image data that is due to limitations in the sensing, signal digitization or data recording process.

OBS

The different sources of this noise could be due to periodic drift or malfunction of the detector, or due to electronic interference among sensor components, or due to problem in the transmission and recording process. The noise, if present in any part of the image can either degrade or totally mask the true radiometric information contained by the digital image. Hence, noise removal is very essential before any preprocessing or analysis of the image could be done.

CONT

Indiana University Laboratory for Spectrochemistry. Plasma Source Mass Spectrometry. "Single-Crystal Scintillator-Based Array Detector for Sector-Field Mass Spectrometry, "[by] David A. Solyom. Beginning with the use of photographic plates in the first quarter of this century, array detectors have had a long history of usage in mass-spectrometric analysis. Array detectors are position-sensitive devices capable of independently measuring ion signals that are incident at different locations on the detector's face. Currently popular among the different types of array detectors available for mass spectrometry are microchannel-plate(s)(MCP) coupled with a phosphor.... The electrons emitted from the rear of the MCP strike the phosphor being held in close proximity(1mm) to the MCP. The phosphor screen is sensitive to the incident electrons and will emit photons in response. The number of emitted photons is proportional to the number of incident electrons and the location on the phosphor from which the photons are emitted coincides with the location of the incident electrons. The photons from the phosphor can then be converted into a proportional electronic signal by an imaging device such as a CCD or CID detector array.

spectrométrie de masse à secteurs à plasma induit (ICP-SFMS).

OBS

The typical particle counter for metering the amount of particulate matter in clean work station air is an optical assembly. Air is drawn into and through the optical subassembly of the particle counter until a pre-set volume of air has been seen. The particles which are drawn into this optical subassembly cause a pulse of light to be scattered for each particle present. The light pulse is sensed by a detector which converts the pulse to an electrical voltage signal. Each signal is cumulatively counted, for the pre-set volume of air. From the amplitude of the voltage, the size of each particle can be determined. All particle counters consist of three basic subassemblies. These are the optical subassembly which "sees" the particles, a pump subassembly which draws the air stream into and through the optical subassembly, and an electronic subassembly for converting each particle size/unit number to an electrical signal. Also available, usually as an option, is some form of printer.
