Recombinant DNA(rDNA) molecules are DNA sequences that result from the use of laboratory methods(molecular cloning) to bring together genetic material from multiple sources, creating sequences that would not otherwise be found in biological organisms. Recombinant DNA is possible because DNA molecules from all organisms share the same chemical structure; they differ only in the sequence of nucleotides within that identical overall structure. Therefore, when DNA from a foreign source is linked to host sequences that can drive DNA replication and then introduced into a host organism, the foreign DNA is replicated along with the host DNA. 2, record 7, English, - recombinant%20DNA

Making more subtle mutations: the "knock in". Although it is often desirable to simply inactivate a gene to determine the importance of a null phenotype, in many cases it is more informative not to inactivate a gene, but rather to modify it so that its function is altered. Again, this is a task (which is the logical equivalent of site directed mutagenesis in a plasmid) that can be solved by homologous recombination. In this case a targeting vector is designed as a replacement vector so that additional genetic sequence are added into the genome. This technique is sometimes called a "knock-in". 2, record 9, English, - replacement%20vector

The technical problems involved in gene transfer therapy make heart transplant surgery look easy. Theoretically, recombinant DNA(inserting strands of DNA where they don’t naturally occur) allows for gene transplantation. Realistically, it is extremely difficult. Researchers have yet to find a way to put a healthy gene into a human somatic cell. DNA is well protected within the cells. DNA writes codes for life, and life would change if DNA were altered. The cell views altered(mutant) DNA as foreign, just like the body views a donor heart. Just like the body has an immune system, the cell has defenses against mutant DNA. 2, record 11, English, - gene%20transplantation

1, record 11, French, - transplantation%20g%C3%A9n%C3%A9tique

This is a system comprised of two enzymatic activities : one a restriction endonuclease that cuts DNA and the other, a methylase that modifies DNA at the same sequence. The idea being that the bacteria's own DNA is modified at the proper sites and hence is protected from the action of the restriction enzyme. However if foreign DNA enters the cell, such as by transformation or by a virus, that DNA is "foreign" and not correctly modified, hence it is a substrate for the restriction enzyme and is cut up into small pieces. 5, record 12, English, - restriction%2Dmodification%20system

The essential ingredients of a recombinant DNA experiment are : 1. A DNA vehicle(vector, replicon) which can replicate in living cells after foreign DNA is inserted into it. 2. A DNA molecule to be replicated(passenger), or a collection of them. 3. A method of joining the passenger to the vehicle. 4. A means of introducing the joined DNA molecule into a host organism in which it can replicate(DNA transformation or transfection). 5. A means of screening or genetic selection for those cells that have replicated the desired recombinant molecule. This is necessary since transformation and transfection methods are inefficient, so that most members of the host cell population have no recombinant DNA replicating in them. This selection or screening for desired recombinants provides a route to recovery of the recombinant DNA of interest in pure form. 1, record 18, English, - DNA%20transformation
