Thermomechanical aqueous/steam treatment (TM-AV). Finely comminuted lignocellulosics, as sawdusts, are prepared as water suspensions (10-15% solids). They are pumped through an homogenizing valve system at temperature ranging from 180 to 240°C. These temperatures are rapidly reached with the injection of steam. The high shear forced on the particles when passing through the homogenizing valves and the flash decompression, after the valves, facilitate the disintegration and solubilization of the hemicelluloses and of about one fifth of the lignin. Thermochemical Fractionation and Liquefaction of Biomass... Proposed three stage biomass utilization scheme.... The first stage(I) is a mild treatment for removal of the hemicellulose fraction. Moderate temperature and pressure are utilized in this stage with the hemicellulose removal either acid or base assisted. The simultaneous isolation, decrystallization and partial saccharification(SIDS) of cellulose is the second stage(II) of the proposed sequence. The second stage reactions are performed at higher temperature and pressures with the intent of obtaining a fermentable oil. The third and final stage(III) is designed to convert lignin to a phenolic oil. Two strategies have been used to clone specific sequences of eukaryotic, genomic DNA in bacteriophage lambda vectors. The first involved cloning of DNA preparations that had been highly enriched for the sequences of interest. Total genomic DNA was digested to completion with a restriction endonuclease and the resulting fragments were separated by preparative gel electrophoresis, reverse-phase column chromatography, or both. The fractions containing the DNA sequences of interest were identified by hybridization to appropriate probes and cloned in bacteriophage lambda vectors. Because these fractionation procedures may yield an overall enrichment of as much as 100-fold to 300-fold, the number of recombinant bacteriophages that had to be generated and screened for the DNA sequences of interest could be significantly reduced.
