A method for in vitro homologous recombination of pools of selected mutant genes by random fragmentation and polymerase chain reaction(PCR) reassembly. Computer simulations called genetic algorithms have demonstrated the importance of iterative homologous recombination for sequence evolution.

As well as addressing the advantages (or lack thereof) of recombination, DNA shuffling provides a powerful tool for improving enzyme function in vitro.

La méthode la plus intéressante du point de vue de l'étude de l'évolution [moléculaire], est celle du DNA shuffling. Dans cette technique, l'ADN d'un gène est tronqué en fragments de 10 à 50 paires de bases, qui sont soumis à des cycles répétés de PCR sans amorce. Les produits PCR résultants sont sélectionnés pour leur expression phénotypique. Ce type d'expérience permet de simuler in vitro l'évolution moléculaire de séquences complexes. En mélangeant l'ADN de deux gênes, la création de chimères recombinantes peut également être envisagée.
