In broad outline the plus-and-minus method calls for synthesizing pieces of DNA that are complementary to various regions of the DNA being sequenced. Because of the way the synthesis is manipulated each piece ends either with or just before a particular, identifiable nucleotide(A, G, T or C). By performing the synthesis to produce a large number of pieces, each one ending with a successive nucleotide in the sequence, it is possible to establish the sequence of the entire DNA molecule.
