Conceivably the individual variations in promoter sequences reflect differences in promoter efficiency, allowing different DNA functional units to be transcribed to lesser or greater degrees. La première méthode utilisée pour repérer la position réelle de différentes mutations du bactériophage lambda est basée sur la possibilité de fragmenter mécaniquement l'ADN en des segments de taille définie. Les fragments qui ont une densité différente peuvent être séparés les uns des autres par centrifugation à l'équilibre dans un gradient de chlorure de césium. Les fragments riches en GC ayant une densité plus grande que les fragments riches en AT, la séparation devient une opération de routine si les différents segments possèdent des rapports AT/GC distincts.
