Liquid gel chromatography(LGC) is an established laboratory technique for the differential fractionation and separation of molecules. The underlying premise for simple LGC is that molecules elute from a gel bed in order of decreasing molecular weight. Generally, gel particles fill a chromatography column to form a bed. When the molecules to be partitioned, such as cells or proteins are introduced and driven through the bed by supplemental eluent, the molecules separate on the basis of molecular weight. 2, record 5, English, - liquid%2Dgel%20chromatography

Of the four basic types of chromatography, the two gas systems are independent and not subject to subdivisions. Thus the terms "gas-liquid chromatography" and "gas-solid chromatography" adequately identify the techniques involved. There are two major limitations for the transferred monolayer systems :(1) Thermodynamic instability renders them suffering from lack of long-term durability;(2) the transferring methods require the supporting substrates to be planar, which will not provide large enough surface area for many important applications, such as enzymatic catalysis. Thus, direct graft of phospholipids to solid substrate to form a highly packed monolayer holds great promise in biomembrane mimetic applications. A method, first reported by Pidgeon and Venkataram to bond covalently phospholipids on a silica gel surface for chromatography applications, has greatly inspired our interest in further developing a stable and robust self-assembled lipid monolayer on solid surfaces to mimic the functionality of biomembranes, especially the surface activation of membrane-bound enzymes by the lipid self-assembled monolayers. 2, record 17, English, - lipid%20monolayer%20on%20silica%20gel

Silica Gel Column Chromatography.... Getting reproducibility when doing silica gel(or alumina) separations with column chromatography can be a problem. If you do not carefully control the water/moisture content of the silica gel and the mobile phase, moisture is added to or removed from the silica gel during the separation. This results in retention times that either increase(with water addition to the silica gel) or decrease(with water loss from the silica gel).... The reseacher attempting to optimize the flash chromatography of new compounds should investigate the effect of moisture on the separation being attempted. Various glass chromatographic columns containing silica gel with different amounts of moisture can be run side by side to see which column produces the optimum results. 1, record 18, English, - silica%2Dgel%20column%20chromatography

Protein purification development and assay : Retentate Chromatography is a new SELDID development which allows rapid protein separation of complex biological fluids on ProteinChip arrays using the bioprocessor accessory for controlled washing of different chromatographically prepared surfaces(anion, cation, IMAV, normal & reversed phase). Retentate mapping’ has many advantages over 2-D gel electrophoresis and other elution chromatography methods for speeding purification protocol development as well as replacing Western Blot, HPLC and ELISA assays. 1, record 24, English, - bioprocessor

1, record 24, French, - processeur%20biologique

Two strategies have been used to clone specific sequences of eukaryotic, genomic DNA in bacteriophage lambda vectors. The first involved cloning of DNA preparations that had been highly enriched for the sequences of interest. Total genomic DNA was digested to completion with a restriction endonuclease and the resulting fragments were separated by preparative gel electrophoresis, reverse-phase column chromatography, or both. The fractions containing the DNA sequences of interest were identified by hybridization to appropriate probes and cloned in bacteriophage lambda vectors. Because these fractionation procedures may yield an overall enrichment of as much as 100-fold to 300-fold, the number of recombinant bacteriophages that had to be generated and screened for the DNA sequences of interest could be significantly reduced. Subsequently, the development of efficient in vitro packaging systems and in situ hybridization techniques eliminated the need to enrich the starting DNA for the sequences of interest. 1, record 27, English, - reverse%20phase%20column%20chromatography
