L'immunodiffusion est utilisée pour étudier les spécificités d'un Ac donné vis à vis de différents Ag (ou vice versa, d'un Ag donné vis à vis de différents Ac) : Antigène Ag et anticorps Ac solubles diffusent radialement, dans un milieu gélosé sérologiquement neutre, à partir de puits creusés dans le gel et où ont été réalisés les dépôts. Les complexes immuns qui se forment éventuellement à la rencontre de l'Ag et de l'Ac sont insolubles et précipitent sous la forme d'arcs d'immunoprécipitation.
