Ionic detergent used for denaturation of protein and in electrophoresis of protein subunits.

Detergent solubilization has been used with great success to study the major proteins in various membranes. Typically, the membranes are first heated to 100°C in a 1% solution of the strong detergent SDS(which disrupts all non-covalent protein-protein and protein-lipid interactions and completely unfolds the proteins), then layered on top of a polyacrylamide gel containing SDS, and lastly subjected to electrophoresis in a strong electric field for several hours. The amount of anionic SDS bound to protein is roughly equal to the mass of the polypeptide, with the result that the negative charge of the detergent overwhelms the intrinsic charge of the protein. Consequently, each individual protein migrates in the electric field at a rate determined by its molecular weight.

Lors de l'électrophorèse en gel filtrant les protéines sont séparées selon leur taille moléculaire (ou encombrement stérique) et leur charge électrique. L'encombrement stérique d'une molécule dépend du poids moléculaire et de la configuration spatiale de la protéine. Pour mesurer le poids moléculaire d'une protéine, il faut donc éliminer l'influence de la charge et de la configuration spatiale. Un agent dénaturant, le dodécyl sulfate de sodium, permet d'obtenir ce résultat. Grâce à son affinité pour les protéines sur lesquelles il se fixe très solidement et en grande quantité, il leur confère une charge électrique constante par unité de surface et modifie considérablement leur configuration spatiale. C'est pourquoi le traitement par le SDS permet de mesurer directement le poids moléculaire d'une protéine après électrophorèse dans un gel de polyacrylamide.

Differential Display(DD)... and a conceptually similar technique, arbitrarily primed PCR fingerprinting of RNA(APPCR) 19, both PCR amplify subsets of an mRNA population and separate the resulting cDNAs by denaturing polyacrylamide gel electrophoresis. As originally described by Patrick O’Farrell for analyzing Escherichia coli proteins in 1975, 2-DGE combines the electrophoretic separation of denatured proteins bysoelectric point charge differences in the first dimension with separation based on molecular size differences in the second dimension. The proteins, which can be detected using protein-specific stains, appear as constellations of spots in the 2-D space of the gel. Over the 20-year history of 2-DGE, numerous algorithms have been developed for comparing 2-DGE patterns and quantitatively analyzing protein abundance.

Starch gel electrophoresis has been a popular technique, although it is now somewhat superseded by the use of polyacrylamide gels, which minimize convection and diffusion effects. A block or "plate" of starch gel is prepared, and the sample is applied in a narrow band(line) across the block about midway between the ends, which are contacted with electrodes through a connecting bridge. When current is passed through the cell, the different components of a mixture move with velocities that depend on their electric charges, their sizes, and their shapes. As electrophoresis proceeds, the negatively charged components migrate toward the anode and the positively charged components migrate toward the cathode. The result is a series of separated bands or lines of sample constituents, such as visualized by a stain.

A technique that has become useful in studies of wheat proteins is called gel electrofocusing. In practice this technique is similar to gel electrophoresis but it differs in principle. It separates a complex mixture of proteins according to a characteristic property called isoelectric point. Proteins that have different amino acid compositions have different isoelectric points. Resolution of wheat proteins by this technique is slightly better than that obtainable with gel electrophoresis, but the albumin(water-soluble) and gliadin(alcohol-soluble) components overlap in their separation range, both having isoelectric points in the region from five to eight on the scale used to express acidity or alkalinity of water solutions(pH).

Two strategies have been used to clone specific sequences of eukaryotic, genomic DNA in bacteriophage lambda vectors. The first involved cloning of DNA preparations that had been highly enriched for the sequences of interest. Total genomic DNA was digested to completion with a restriction endonuclease and the resulting fragments were separated by preparative gel electrophoresis, reverse-phase column chromatography, or both. The fractions containing the DNA sequences of interest were identified by hybridization to appropriate probes and cloned in bacteriophage lambda vectors. Because these fractionation procedures may yield an overall enrichment of as much as 100-fold to 300-fold, the number of recombinant bacteriophages that had to be generated and screened for the DNA sequences of interest could be significantly reduced. Subsequently, the development of efficient in vitro packaging systems and in situ hybridization techniques eliminated the need to enrich the starting DNA for the sequences of interest.
