Detergent solubilization has been used with great success to study the major proteins in various membranes. Typically, the membranes are first heated to 100°C in a 1% solution of the strong detergent SDS(which disrupts all non-covalent protein-protein and protein-lipid interactions and completely unfolds the proteins), then layered on top of a polyacrylamide gel containing SDS, and lastly subjected to electrophoresis in a strong electric field for several hours. The amount of anionic SDS bound to protein is roughly equal to the mass of the polypeptide, with the result that the negative charge of the detergent overwhelms the intrinsic charge of the protein. L'encombrement stérique d'une molécule dépend du poids moléculaire et de la configuration spatiale de la protéine. Pour mesurer le poids moléculaire d'une protéine, il faut donc éliminer l'influence de la charge et de la configuration spatiale. Un agent dénaturant, le dodécyl sulfate de sodium, permet d'obtenir ce résultat. Grâce à son affinité pour les protéines sur lesquelles il se fixe très solidement et en grande quantité, il leur confère une charge électrique constante par unité de surface et modifie considérablement leur configuration spatiale. One current proteomic tool for visualizing and quantitating all proteins expressed in a ological system at a given time is two-dimensional gel electrophoresis(2-DGE). As originally described by Patrick O’Farrell for analyzing Escherichia coli proteins in 1975, 2-DGE combines the electrophoretic separation of denatured proteins bysoelectric point charge differences in the first dimension with separation based on molecular size differences in the second dimension. A technique that has become useful in studies of wheat proteins is called gel electrofocusing. In practice this technique is similar to gel electrophoresis but it differs in principle. Two strategies have been used to clone specific sequences of eukaryotic, genomic DNA in bacteriophage lambda vectors. The first involved cloning of DNA preparations that had been highly enriched for the sequences of interest. Total genomic DNA was digested to completion with a restriction endonuclease and the resulting fragments were separated by preparative gel electrophoresis, reverse-phase column chromatography, or both. The fractions containing the DNA sequences of interest were identified by hybridization to appropriate probes and cloned in bacteriophage lambda vectors. Because these fractionation procedures may yield an overall enrichment of as much as 100-fold to 300-fold, the number of recombinant bacteriophages that had to be generated and screened for the DNA sequences of interest could be significantly reduced.
