An analytical or separation procedure similar to gel electrophoresis. It is used to separate proteins and other charged molecules on the basis of their isoelectric point. A series of ampholytes are used to establish a pH gradient within a support gel. Méthode de séparation très résolutive des protéines qui repose sur la distinction de ces macromolécules selon leur point isoélectrique (pHi). Chaque protéine en effet, comme n'importe quel ampholyte, est définie par son point isoélectrique, valeur du pH du milieu pour lequel sa charge électrique résultante est nulle. Placée à un pH supérieur à son pHi la protéine se charge négativement par perte de protons. A un pH inférieur au pHi, la protéine se charge positivement par capture de protons. One current proteomic tool for visualizing and quantitating all proteins expressed in a ological system at a given time is two-dimensional gel electrophoresis(2-DGE). As originally described by Patrick O’Farrell for analyzing Escherichia coli proteins in 1975, 2-DGE combines the electrophoretic separation of denatured proteins bysoelectric point charge differences in the first dimension with separation based on molecular size differences in the second dimension. The proteins, which can be detected using protein-specific stains, appear as constellations of spots in the 2-D space of the gel. A technique that has become useful in studies of wheat proteins is called gel electrofocusing. In practice this technique is similar to gel electrophoresis but it differs in principle. It separates a complex mixture of proteins according to a characteristic property called isoelectric point. Proteins that have different amino acid compositions have different isoelectric points. Resolution of wheat proteins by this technique is slightly better than that obtainable with gel electrophoresis, but the albumin(water-soluble) and gliadin(alcohol-soluble) components overlap in their separation range, both having isoelectric points in the region from five to eight on the scale used to express acidity or alkalinity of water solutions(pH). 2, record 5, English, - gel%20electrofocusing
