The results indicate that arsenite alone is mutagenic in human cells using the supF reporter gene. The pZ189 shuttle vector may provide a model to study the molecular nature of co-mutagenesis of arsenic and other environmental agents. Further characterization of arsenic' s effects on DNA repair and mutational spectra may be useful in the development of molecular markers in studies of arsenic carcinogenesis in human populations. 2, record 1, English, - shuttle%20vector

Recombinant techniques... allow saturation mutagenesis of selected regions or sites of a gene. By these techniques, families of related enzymes are produced in a single cloning exercise. A population of bacterial colonies so produced will carry the mutant genes; each colony will harbor the gene coding for a single recombinant enzyme. Screening techniques may allow the colonies to be rapidly examined for mutant protein activity. Only the subset of colonies containing active enzymes will require further characterization. A comparison of mutant and wild-type proteins(or genes) will reveal sites that modulate activity or function. 1, record 2, English, - recombinant%20enzyme

Gene characterization, What does it mean to characterize a gene at the molecular level? There is no strict definition, but at the minimum it means to define that region of a chromosome, or a transcribed product, that is necessary to encode a specific product or function. To utilize a cloned gene, more information than this must be obtained. To utilize a cloned gene, one needs to know several types of information... 1, record 4, English, - gene%20characterization

Short tandem repeat polymorphisms, also referred to as variable number of tandem repeats(VNTRs), are very important to scientists, because they can be used to identify individuals genetically. Marker systems are used to detect loci that respond to restriction enzyme cleavage by producing a fragment that can have many different lengths. This polymorphism is due to variation in the number of tandem repeats. VNTRs can be identified in various ways. Probes can be developed which will hybridize to complementary sequences after RFLP has been performed on a sample of genomic DNA(Southern blots). A second method involves the use of PCR. If the sequence surrounding the VNTR is known, PCR primers(oligonucleotides) can be designed to amplify the repeat region(s). The information obtained from these experiments is useful in the fields of forensics, population characterization, human gene mapping, identification of genes involved in disease, relationship testing, and identifying genes of interest such as those that encode proteins containing zinc finge. 3, record 5, English, - variable%20number%20of%20tandem%20repeats
