Both cyclases are activated by the host calmodulin. Their genes have been cloned and expressed in E. coli... using an original technique, using E. coli cells expressing one of the components of the activation system for the screening of a library of the pathogen's genome. Accordingly, this technique must be considered as the first example of the "double hybrid" technique : it has for example been used to clone the cDNA for calmodulin from human or mouse cell lines, by functional complementation of the B. pertussis adenylyl cyclase gene.

Chromogranin B(secretogranin I or Scg-1) is a tyrosine-sulfated secretory protein found in a wide variety of peptidergic endocrine cells. Benedum et al.(1987) isolated a 2. 5-kb cDNA clone from a cDNA library of human pheochromocytoma. Chromogranin B is a 657 amino acid long polypeptide of 76 kilodaltons and is preceded by a cleaved end-terminal signal peptide of 20 residues. Chromogranin B was assigned to chromosome 20 by Craig et al.(1986). Craig et al.(1987) showed by in situ hybridization that the CHGB locus is on 20pter-p12. Jenkins et al.(1991) mapped the murine gene, symbolized Scg-1, to chromosome 2 by in situ hybridization and by interspecific backcross analysis.

Chromosome walking, one strategy for filling in gaps, involves hybridizing a primer known sequence to a clone from an unordered genomic library and synthezing a short complementary strand(called walking along a chromosome). The complementary strand is then sequenced and its end used as the next primer for further walking; in this way the adjacent, previously unknown region is identified and sequenced. Chromosome walking is also used to locate specific genes by sequencing the chromosomal segments between markers that flank the gene of interest.
