Homologous recombination is that which we are taught about in introductory biology class. It is the crossing-over process observed during meiosis, though it occurs in bacteria as well. The process involves pairing up of similar DNA sequences, with then one sequence replacing the other, or indeed for meiotic crossing over, with the generation of now two recombinant strands, one for each chromosome homolog, rather than just one. In either case, key is that it is DNA that is highly similar in terms of DNA sequence that is being either replaced or swapped. This allows for a replacement of alleles following horizontal gene transfer, or allele swapping... 2, record 1, English, - homologous%20recombination

1, record 2, Spanish, - gen%20desactivado

Making more subtle mutations : the "knock in". Although it is often desirable to simply inactivate a gene to determine the importance of a null phenotype, in many cases it is more informative not to inactivate a gene, but rather to modify it so that its function is altered. Again, this is a task(which is the logical equivalent of site directed mutagenesis in a plasmid) that can be solved by homologous recombination. In this case a targeting vector is designed as a replacement vector so that additional genetic sequence are added into the genome. This technique is sometimes called a "knock-in". 2, record 5, English, - replacement%20vector

Analysis of the surface of the crystallographic models of most proteins should target at least one surface residue whose replacement would substantially increase the stability of the protein.... Unfortunately, most enzymes do not presently have a crystallographic model available for identification of one or more residues having exposed apolar side chains.... For investigators having neither a crystallographic model of the enzyme of interests or a facility for obtaining and engineering the structural gene, use of a cross-linking reagent is recommended to increase stability. This approach is based on diminishing the polypeptide entropy that is the principal thermodynamic quantity stabilizing the denatured form. 2, record 6, English, - crystallographic%20model
