Today, the relative ease of gene sequencing is leading to an explosion in the derivation of protein primary structure by direct gene translation. This information is being stored in DNA databanks. The current problem with the design of enzymes is that we are still unable to predict reliably the three-dimensional structure adopted by a known primary structure(amino acid sequence)... Minor changes in primary structure can result in the formation of additional intramolecular intersubunit electrostatic or hydrophobic bonds. Even a small number of these bonds can drastically increase the stability of subunit enzymes against dissociative inactivation.

What is Post-Genomics Biology? In this era of whole-genome sequencing, the questions are changing-the driving force is no longer to clone and sequence genes in the random fashion that was available before, but rather to use the wealth of information available from the sequencing projects to understand gene function and gene product interactions at the physiological level. Do homologous proteins have homologous functions in different organisms? How do we use information from a 'simple’ organism to focus research in more complex ones? What do all these genes do that have not been seen using traditional methods? How can we understand the different roles of members of gene families which are only now appearing as a result of the genome sequencing projects?

Chromosome walking, one strategy for filling in gaps, involves hybridizing a primer known sequence to a clone from an unordered genomic library and synthezing a short complementary strand(called walking along a chromosome). The complementary strand is then sequenced and its end used as the next primer for further walking; in this way the adjacent, previously unknown region is identified and sequenced. Chromosome walking is also used to locate specific genes by sequencing the chromosomal segments between markers that flank the gene of interest.
