Du Pont and Duke University collaborators have developed the Biolistic gene gun, a particle delivery system which employs microcarriers coated with genetic material to insert DNA into animal cells. The microcarriers are accelerated on macrocarriers in a stream of helium to fire the microcarriers into external tissue or, using surgery, into the livers of mice. The system has been used to insert genes encoding the enzyme, firefly luciferase, into mice and the next stage is likely to be insertion of a therapeutically useful gene into animal liver cells which would show the procedure to be of potential use in gene therapy. 1, record 1, English, - gene%20gun

The technical problems involved in gene transfer therapy make heart transplant surgery look easy. Theoretically, recombinant DNA(inserting strands of DNA where they don’t naturally occur) allows for gene transplantation. Realistically, it is extremely difficult. Researchers have yet to find a way to put a healthy gene into a human somatic cell. DNA is well protected within the cells. DNA writes codes for life, and life would change if DNA were altered. The cell views altered(mutant) DNA as foreign, just like the body views a donor heart. Si on parvient par exemple à introduire un gène d'une cellule humaine dans l'ADN d'une bactérie, ce gène se comportera dans cette bactérie comme chez l'homme. Pour pouvoir réaliser une telle «transplantation génétique» il faut que le gène en question soit découpé au bon endroit de la chaîne d'ADN de la cellule humaine et que l'ADN récepteur de la bactérie puisse être ouvert artificiellement au bon endroit. Pour faire sortir les gènes, on fait intervenir des enzymes dites de restriction et des ligases (enzymes collantes) pour leur incorporation dans le nouvel ADN. Si on parvient par une telle transplantation de gène à faire en sorte que la cellule hôte produise la protéine voulue, commandée par le gène du donneur, on obtiendra alors une protéine recombinante. La cellule hôte, une bactérie dans notre exemple, se multiplie rapidement avec son nouveau gène, ce qui fait que très rapidement, une multitude de bactéries seront en mesure de produire la protéine voulue en grande quantité, ce qui n'aurait pas été possible avec la cellule donneuse. Les bactéries sont particulièrement indiquées, car une partie de leur ADN se trouve sous forme de plasmide. Of all the therapies that pharmacologists see on the horizon, perhaps the most tantalizing are ones that would cure disease by fixing damaged genes. Diseases such as cystic fibrosis, hemophilia, and severe combined immunodeficiency are termed "single-gene" disorders because a single abnormal gene makes an abnormal protein, which, in turn, results in the sumptoms of the disease. In theory, "genetic surgery" could cure these single-gene diseases by cutting out the abnormal gene and replacing it with a working one.
