Intravitreal gene therapy represents an approach to treating retinal diseases by delivering therapeutic genes directly into the vitreous humor of the eye. This method uses a viral vector, often an adeno-associated virus(AAV), to carry genetic material into retinal cells. Once inside, the therapeutic genes are expressed to address genetic deficiencies or modify biological pathways, offering a long-term or potentially permanent treatment... for conditions like wet age-related macular degeneration(AMD), diabetic macular edema, and inherited retinal dystrophies. 2, record 2, English, - intravitreal%20gene%20therapy

... the suprachoroidal injection approach delivers the gene therapy payload via the potential space between the sclera and the choroid(suprachoroidal space, or SCS). This is a potentially favored injection site for gene therapy because it may reduce the likelihood of eliciting an immunologic response due to its unique location with respect to the blood-retina barrier while simultaneously permitting adequate gene expression to control the nAMD [neovascular age-related macular degeneration] disease process. Surgical access to the SCS can be facilitated by creation of a sclerotomy, followed by introduction of a catheter or cannula. Separately, and potentially of greater interest, is for suprachoroidal gene therapy to be delivered in the clinic setting by utilizing hypodermic needles or hollow microneedles. 1, record 3, English, - suprachoroidal%20gene%20therapy

There has been interest in applying nanogels to drug delivery systems, such as protein delivery and gene delivery. In general, chemically cross-linked nanogels are synthesized by microemulsion polymerization or a crosslinking reaction of intra-associated polymer molecules. We develop tailor-made functional nanogels to create novel nanobiomaterials(nanogel engineering) by the self-assembly of functional associating polymers as building blocks. In particular, nanogels of cholesteryl group-bearing pullulan(CHP) selectively interact with proteins as a host and are useful as artificial molecular chaperone and drug carriers such as cancer immune therapy. 3, record 8, English, - nanogel

Du Pont and Duke University collaborators have developed the Biolistic gene gun, a particle delivery system which employs microcarriers coated with genetic material to insert DNA into animal cells. The microcarriers are accelerated on macrocarriers in a stream of helium to fire the microcarriers into external tissue or, using surgery, into the livers of mice. The system has been used to insert genes encoding the enzyme, firefly luciferase, into mice and the next stage is likely to be insertion of a therapeutically useful gene into animal liver cells which would show the procedure to be of potential use in gene therapy. 1, record 9, English, - gene%20gun

The technical problems involved in gene transfer therapy make heart transplant surgery look easy. Theoretically, recombinant DNA(inserting strands of DNA where they don’t naturally occur) allows for gene transplantation. Realistically, it is extremely difficult. Researchers have yet to find a way to put a healthy gene into a human somatic cell. DNA is well protected within the cells. DNA writes codes for life, and life would change if DNA were altered. The cell views altered(mutant) DNA as foreign, just like the body views a donor heart. Just like the body has an immune system, the cell has defenses against mutant DNA. 2, record 17, English, - gene%20transplantation

1, record 17, French, - transplantation%20g%C3%A9n%C3%A9tique

Understanding cryptochrome’s connection to the body’s internal clock could change how doctors treat certain illnesses. Sleep irregularities as temporary as jet lag and as serious as insomnia could eventually be addressed with a better understanding of the circadian rhythm. Naturally occurring virus releases its genetic material into cells. Whether or not the genes become integrated into the DNA or the infected cell, they soon direct the synthesis of new viral particles that injure the cell and infect others. To convert a wild-type virus into a safe gene therapy vector, scientists replace viral genes with ones specifying therapeutic proteins, while ideally leaving only the viral elements needed for gene expression. Such vectors should enter cells and give rise to helpfull proteins but should not multiply. 1, record 34, English, - gene%20therapy%20vector

Nodavirus RNA Replication. We have constructed DNA plasmids and vaccinia virus recombinants that express the RNA replicase of a nodavirus. In the cytoplasm of transfected or infected mammalian cells, this enzyme selectively amplifies its own mRNA by RNA replication. In 1994, the Presidents of the Medical Research Council of Canada(MRC), the Natural Sciences and Engineering Research Council of Canada(NSERC), and the Social Sciences and Humanities Research Council of Canada(SSHRC), collectively appointed a multi-disciplinary working group(the Tri-Council Working Group) and gave it the mandate to develop new policies and regulations to replace the Councils’ existing guidelines for research involving humans. The "Code of Ethical Conduct for Research Involving Humans"(the Code) is the result of that process and supersedes both the MRC "Guidelines on Research Involving Human Subjects" 1977 and the 1987 SSHRC "Ethics Guidelines for Research with Human Subjects". This Code complements the 1990 MRC "Guidelines for Research on Somatic Cell Gene Therapy in Humans". 1, record 36, English, - Code%20of%20Ethical%20Conduct%20for%20Research%20Involving%20Humans

For gene therapy to realize its clinical potential, there must be efficient and safe strategies of delivering therapeutic genes to somatic cells in vivo. Perhaps this problem simply represents a special case of drug delivery in which the therapeutic gene constitutes the drug. But the relevance of traditional drug development to technological developments in gene therapy is questionable. Recombinant genes that can independently express a therapeutic RNA or protein are extraordinarily large and polar molecules--10, 000 times larger than traditional pharmaceutical agents. Moreover, the administration of genes as therapy requires vehicles--so-called vectors--that encapsulate the gene and guide it to the target cell. The binding of the gene to the cell, its internalization, the transport of the administered genome to the nucleus, and the expression of the gene all constitute potential limitations of this process. 1, record 37, English, - therapeutic%20gene

The Goal of Gene Therapy for Cystic Fibrosis... Gene therapy offers the best hope for a life-saving treatment which tackles the root cause of cystic fibrosis(CF), rather than only treating the symptoms... These early trials test the safety and efficiency of the adenovirus delivery system. Patients are monitored to detect whether the normal CF gene has turned on. If so, the gene will produce a normal protein that is vital to the health of cells lining the respiratory tract. 3, record 39, English, - Cystic%20Fibrosis%20gene

1, record 40, French, - g%C3%A8ne%20consid%C3%A9r%C3%A9%20comme%20m%C3%A9dicament

Virosome Vectors for gene therapy. The long term goal of this work is to develop a targeted, efficient, liposome-based vector for the delivery and insertion of the CFTR gene to lung cells. To do this, a combination of liposometechnology and molecular biology will be used. Three observations have led us to propose the outlined research :(i) The efficiency of liposome fusion to cells or organelles can be greatly enhanced by incorporating viral fusion proteins intovesicles.(ii) After liposome-cell fusion, the contents of the liposomes are released into the cell' s cytoplasm.(iii) By delivering DNA to nonlysosomal compartments of eukarvotic cells the efficiency of transfection can be greatly enhanced... 1, record 41, English, - virosome
