The human immunodeficiency virus (HIV) genome encodes at least seven groups of viral proteins. These groups of proteins include the three classes of polypeptides present in the majority of animal retroviruses: 1. structural, nonenvelope polypeptides encoded by the gag gene; 2. enzymes required for virion replication (reverse transcriptase/r) and for cleavage of viral precursor proteins (protease/pr), both encoded by the pol gene; 3. envelope polypeptides encoded by env gene. In addition, HIV’s possess genes that encode four sets of polypeptides not found in the majority of typical retroviruses. These include a transactivating regulator of RNA translation (tat), a cis-acting downregulator of RNA transcription (nef), a regulator that modulates the expression of structural proteins (rev), a protein that modulates viral infectivity (vif), and two proteins (vpr,vpu) whose functions are still unclear. La fixation du V.I.H. à la cellule CD4+. La fixation de la particule virale sur la cible cellulaire se fait par interaction de la gp120 et de la molécule CD4, ce qui découvre la glycioritéine transmembranaire gp41. Une des extrémités de la gp41 s'introduit alors dans la membrane cellulaire et cet accrochage engendre la fusion entre la membrane virale et la membrane cellulaire. Le mécanisme de la pénétration intracellulaire du V.I.H. reste encore discuté. L'enveloppe virale conditionne donc l'infectivité du V.I.H. puisqu'il s'attache spécifiquement à la cellule qu'il infecte par l'intermédiaire de ses glycoprotéines d'enveloppe. The coat of the HIV virion bears numerous viral proteins "spikes" that project into the external medium. Each spike probably consists of four molecules of a protein called gp120 on the outside and the same number of gp41 embedded in the membrane. These envelope proteins play a crucial role when HIV binds to, and enters target cells. Underneath the envelope is a layer of matrix protein called p17, which in turn surrounds the core, or capsid.
