Affinity elution is complementary to affinity chromatography in that in the former technique the specificity of interaction is at the stage of desorption from the chromatographic support material, whereas in the latter the specificity occurs at the stage of adsorption. Often, all substances are not eluted from the column with a single solvent. After elution of part of the solutes, a different solvent or solvent mixture is added that shows a greater eluting power for the remaining solutes.
