A gradient method is recommended for samples that cannot be easily separated by isocratic methods because of their wide k range. In gradient elution the eluent strength is increased during the separation by changing the composition of the mobile phase. As a result, the analysis time is reduced and the quality of the separation is improved as well as the detection limit.

An affinity purification technique based on ’non-specific' adsorption to an ion exchanger. Cation exchangers appear to be particularly useful for this purpose. Purification specificity is achieved using gradient elution with substrate.

Affinity elution is complementary to affinity chromatography in that in the former technique the specificity of interaction is at the stage of desorption from the chromatographic support material, whereas in the latter the specificity occurs at the stage of adsorption. In affinity elution the enzyme of interest (along with others) is adsorbed onto an ion exchanger ... and then is eluted specifically by the appropriate substrate.

After elution of part of the solutes, a different solvent or solvent mixture is added that shows a greater eluting power for the remaining solutes. In solvent gradient analysis, mixed solvents are used, but their volume ratios are varied continuously throughout the elution, usually in a linear fashion.

After elution of part of the solutes, a different solvent or solvent mixture is added that shows a greater eluting power for the remaining solutes... a concentrated salt or other reagent in the eluting solvent may be continuously diluted, as in salt gradient analysis. The salt may act as a salting-out agent(precipitating) for the solutes, and the salting-out ability decreases continuously.
