DNase Agar. DM132 A medium for the presumptive identification of pathogenic staphylococci by demonstration of DNase production.

Deoxyribonuclease(DNAse) agar actually has DNA in it. Deoxyribonuclease is an exoenzyme excreted from the cell which will break the DNA down into smaller molecules. There are 2 kinds of DNAse agar that can be used. 1) The first medium has no indicator, but is does require HCl(hydrochloric acid) reagent to be added after growth has occurred on the plate. Without DNAse enzyme, the HCl precipitates out the undestroyed DNA. The rest of the agar with the remaining DNA, which binds to HCl will have turned cloudy. Around the growth area, if the bacterium has made DNAse and destroyed the DNA, there will be clear zones seen. 2) DNAse agar with the indicator methyl green can be used. In this medium, when the DNA in the medium is hydrolyzed by the bacterial DNAse enzyme, the pieces of cut DNA release from binding to methyl green, and you will see a yellowish/orange zone around the growth area. OBJECTIVE : To identify the presence of the DNAse enzyme.

Cet acide mixte [...] a des propriétés particulières dépendantes de l'action combinée de ses deux bases acidifiables, et nous avons cru, par cette raison, devoir lui conserver un nom particulier. Celui d'acide nitro-muriatique nous a paru le plus convenable.

Here, wood chips are impregnated with 30% hydrochloric acid in a mixer and then transferred by a conveyor into the reactor. Because the chips are not predried, the acid concentration in them is reduced to 20%. The reactor consists of a hexagonal upright vessel equipped with numerous inclined trays. The impregnated wood is moved from the top toward the bottom by means of a vibrator and thus the prehydrolysis is completed after 45 minutes. The acid, containing chiefly pentoses, is filtered off and part of it is returned to the mixer. In this way, the sugar concentration in the prehydrolyzate is increased to about 20% to 25%. In a second step, the material, which has again been mixed with 30% hydrochloric acid, is passed through the reactor and hydrogen chloride gas is injected at the bottom in a countercurrent. In this way, the acid concentration is increased to 40% to 41% or more. The hydrolyzed material is conveyed from the bottom of the reactor to a drying chamber and is dried there with anhydrous hydrogen chloride gas which has previously been passed through a calcium chloride solution. The hydrolyzed material, consisting of lignin and sugar, is extracted with water and the sugar solution is subjected to a posthydrolysis.

The etched surface is now completely dried(...) and placed horizontally on plasticene with the polished surface up. As soon as the surface is more or less ’room dry’, acetone is carefully poured over the surface and continually squirted over the surface with a small pipette. The acetate sheet is immediately rolled on during this process.(...) The acetate sheet is pressed down with the finger and gently held down for about 5-10 minutes so that the peel can shrink. This shows itself in a slight waviness of the surface. The film is then carefully lifted off.

"Technique des pellicules" La surface aplanie d'une roche est attaquée à l'acide chlorhydrique dilué pendant quelques minutes, lavée et séchée. Une empreinte en est prise au moyen d'une pellicule, dont il existe différentes formules. Le film, arraché de la roche, donne une réplique de la surface. Cette technique rapide et peu coûteuse donne parfois des résultats supérieurs à la plaque mince.
