Liquid gel chromatography(LGC) is an established laboratory technique for the differential fractionation and separation of molecules. The underlying premise for simple LGC is that molecules elute from a gel bed in order of decreasing molecular weight. Generally, gel particles fill a chromatography column to form a bed. When the molecules to be partitioned, such as cells or proteins are introduced and driven through the bed by supplemental eluent, the molecules separate on the basis of molecular weight.

Due to the drop in pressure that occurs in high-pressure liquid chromatography(HPLC) as the solvent passes from the column to the detector, any dissolved gases(most commonly air) are partially released as bubbles within the detector, producing anomalous and misleading results. To avoid this problem, solvents are degassed before use, either by a nitrogen purge or by heating and stirring under vacuum.

The initial solubilization of yolk platelets demonstrated that the lipid was present in the soluble fraction containing lipovitellin and phosvitin. These two proteins have been resolved and the lipovitellin fraction was shown to exhibit the absorption maximum at 375 nm that characterizes DCD [dorsalizing cytoplasmic determinant]. The lipovitellin was purified and its total lipids extracted and separated on HPLC [high-performance liquid chromatography]. A lipid was found that has identical elution properties from a C18 reverse HPLC column and mass as DCD. Therefore, lipovitellin, a zinc protein known to contain 17% lipid by weight, is the oocyte storage protein for DCD.

A chromatographic technology used to separate and quantitate mixtures of substances in solution.

The sample is injected into a moving stream of solvent, which flows through a column and detector. The passage through the column separates the sample compounds by one of four different methods: adsorption, partition, ion exchange, or size exclusion. The solvent is pumped through the system at a controlled pressure, which can be varied between 500 and 5000 psi. With the particular column and elution method used for the assay, the elution times relative to the internal standard characterize the compounds and are used to identify them. The peak areas relative to the internal standard are proportional to their concentration in the sample and are used to quantitate them.

A special form of liquid chromatography, high-performance liquid chromatography(HPLC), has become especially important for the separation of complex mixtures of nonvolatile materials. Essentially, it involves the use of very small particle size in column packing, with high pressure to increase the rate of flow and shorten the separation time to a few minutes.

One of the most significant advances in chromatography in recent years has been the introduction of high performance liquid chromatography. Originally intended for the separation of small organic molecules that were soluble in non-aqueous solvents, the technique rapidly developed into a form suitable for the separation of larger, biological compounds soluble in aqueous solvents. In recent years, following the development of macroporous supports, the technique has become applicable to molecules as large as proteins and nucleic acids.

The term high pressure liquid chromatography (now obsolete) was used in the early stages of development of the technique since the quality of the columns was such that high pressures were needed to force the liquid through.

L'appareil est constitué d'une colonne en acier inoxydable d'une longueur de l'ordre de 200 mm et de diamètre intérieur de 4 mm, remplie de particules de taille inférieure à 20 mm; cette colonne est précédée d'une pompe à haute pression, permettant d'injecter le solvant sous une pression de plusieurs dizaines de bars. Des microvannes assurent l'introduction de quelques microlitres de solution à analyser, en tête de colonne.

An injection port is necessary in all g. c. [gas chromatography] and h. p. l. c. [high-performance liquid chromatography] systems in which syringe injection is employed. The injection port joins the top of the column to the mobile phase supply line, and has a third outlet through a small orifice covered by a self-sealing septum. Injection of samples is done by inserting the hypodermic syringe containing the sample through this septum so that the point of the needle is immediately above the head of the column. Design of the injection port must be such that it can be maintained at a uniform temperature and will possess a very small dead volume.

A device for the extremely sensitive measurement of the refractive index of liquids; used as a detector in liquid chromatography.

The most universal continuous detector for liquid chromatography is the differential refractometer. This detector is universal only to the extent that one can select an eluent the refractive index of which differs from that of the solutes. Pure eluent flows through one cell of the detector while column effluent flows through the other cell. When a solute with a refractive index different from that of the solvent enters the detector, an imbalance occurs, which is transmitted to the recorder. The differential refractometers are relatively insensitive to trace and minor components, and detector stability is greatly affected by small temperature and flow fluctuations, thus requiring very good flow and temperature controls.
