The general principles of chemical-reaction kinetics apply to enzyme-catalyzed reactions, but they also show a distinctive feature not usually observed in nonenzymatic reactions, saturation with substrate.... At a low substrate concentration, the initial reaction velocity vo is nearly proportional to the substrate concentration, and the reaction is thus approximately first order with respect to the substrate. However, as the substrate concentration is increased, the initial rate increases less, so that is is no longer nearly proportional to the substrate concentration; in this zone, the reaction is mixed order. A form of the Michaelis-Menten equation in which reciprocals of both sides are equated to give the linear form, l/V=(Km/Vmax)(1/(S] + 1/Vmax, where V is the reaction velocity,(S) is the substrate concentration, and Km is the Michaelis-Menten constant. Several other linearized forms of the Michaelis-Menten equation have been derived:[S)/V) = (1/Vmax) (S) + K(subscript m]/Vmax) (Hanes Plot), and (V) = (-V/(S] (Km) + (Vmax) (Eadie-Hofstee Plot). The rate of an enzyme reaction(V) varies with the substrate concentration, increasing with an increased concentration until a maximum value(Vmax) is reached. At this point, all enzyme molecules are fully occupied and the rate of reaction is limited by the rate of the catalytic process on the enzyme surface. For most enzymes, the concentration of substrate at which the reaction rate is half maximal(Km) is a measure of how tightly the substrates are bound, with a large value of Km representing a weak binding, and vice versa. 3, record 3, English, - concentration

Il existe des réactions d'ordre zéro, du premier ordre, du deuxième ordre et du troisième ordre, suivant la nature de la relation entre la vitesse de la réaction et la concentration d'un des réactifs dans des conditions données. La vitesse de certaines réactions chimiques est indépendante de la concentration des réactifs : on les appelle réactions d'ordre zéro. Les réactions du deuxième ordre sont celles dont la vitesse est proportionnelle au produit des concentrations de deux réactifs différents, ou au carré de la concentration d'un seul réactif.
