The analytical electron microscope(AEM) is based on a conventional 200 keV transmission electron microscope(TEM) which has been augmented to include the functions of a scanning transmission electron microscope(STEM) and the scanning electron microscope(SEM). Since the electrons must penetrate through the specimen, the specimen dimensions must be no greater than 1 micrometer thick, and preferably in the range of 100 nm. Bulk specimens may be thinned to this range by a combination of mechanical, chemical, and ion sputtering techniques, all of which are available in the microanalysis laboratory. As an imaging tool, the AEM is capable of 0. 3 nm spatial resolution in the TEM mode, 1 nm resolution in the STEM mode, and 3 nm resolution in the SEM mode. Elemental microanalysis is carried out by means of energy dispersive x-ray spectrometry(EDS) and electron energy loss spectrometry(EELS), which together provide sensitivity to all elements with atomic number > 2. In the last 10-15 years many analytical advances in X-ray fluorescence analysis(XRF) have taken place, giving rise to non-destructive ultrasensitive surface analyses of materials. One of the variants of XRF developed is micro-XRF(5-XRF), which is able to analyse the distribution of major, minor and trace elements in microscopic sample surface areas. La micro-fluorescence X est une technique qui consiste à exciter par un micro-faisceau de rayons X les seuils d'absorption des éléments chimiques présents dans un échantillon. La désexcitation de ces atomes induit l'émission de photons dont l'énergie est caractéristique de l'élément en question. Ces photons sont collectés par un détecteur et classés dans des canaux en fonction de leur énergie. On obtient ainsi ce qu'on appelle un spectre d'émission de fluorescence. Ce spectre comporte des pics (ou raies) qui caractérisent les différents éléments présents dans l'échantillon. Tout l'échantillon est balayé point par point par le micro-faisceau et un spectre d'émission est acquis pour chaque point.
