The cameras carrying the photographs are provided with objective lenses identical with those used in the surveying... cameras, and each of them carries in front of its own objective, rigidly fixed thereto, a plane glass plate inclined at an angle of 45° with respect to the optical axis of the said camera, so that said plate... acting as a mirror, produces a virtual image of the camera located at the same side as the mark and in fixed relation to the real camera carrying the photograph.

A plane glass plate between the objective and the object, tilted 45 degrees with respect to the optic axis, can degrade the resulting image. For example: If the object is a white square, then the image appearing beyond the tilted plane glass will have a red edge along one side and a violet edge along the opposite side.

[The Ronchi test] uses a grating of fine parallel lines to test the deviation of a mirror from its correct figure. The grating is put in front of the eye after removing the eyepiece and pointing at a star. The lines should appear straight for a spherical mirror.

More efficient than the Foucault knife-edge test, [the Ronchi test] examines curved mirrors by using a transmission grating with 40 to 200 lines per inch in place of the knife-edge, and a section or slit of this grating in place of the pinhole source.

A Ronchi grating is used as a transparent plate ruled with black lines and equal, clear spaces. It is used as a multiple knife-edge for testing a concave mirror.

La qualité de la surface réflectrice a été testée à l'aide du classique test de Foucault. [...] L'observation d'une surface polluée d'ondes avec un test plus évolué, un test de Ronchi par exemple, donne lieu à une trop grande densité d'information, ce qui devient presqu'impossible à interpréter.

L'inconvénient [du test de Ronchi] est son manque de précision. Contrairement au test de Foucault, le test de Ronchi n'est pas pratiqué au foyer de l'instrument. Pour gagner en précision, il faut d'une part augmenter le nombre de traits au mm [de la trame] et se rapprocher du foyer. Dans ce cas les phénomènes de diffraction sont prépondérants et rendent l'interprétation de l'image impossible. On ne peut donc pas estimer de façon sûre et quantitative une aberration sur le front d'onde avec ce genre de test.

The Molecular Dynamics Sarastro 2000 has a simple light path that makes it ideal for conversion to a 2-photon scope. We retained the ability to use the microscope in the 1-photon, confocal mode by replacing mirror 2 with a beamsplitter that passes IR from the Mira 900 or reflects visible excitation from the on-board 25 mW argon-ion laser. Mirror 3 and the two scanning mirrors were replaced with broad-band mirrors for better IR throughput. We installed a plate that blocks the IR laser beam whenever the lid is opened to change filters, for safety. That is it--No more modifications necessary! To use in the 2-Photon mode, the on-board laser is turned off, the Mira 900 IR laser is turned on, the primary dichroic wheel is rotated to a 680 SP(Chroma) mirror, and the excitation filter and aperture wheels are rotated to the "open" settings. To improve rejection of excitation light, a barrier filter(680 SP, Chroma) is used in the detector housing.

Deux photons peuvent combiner leurs énergies pour provoquer le passage vers un état excité plus élevé du fluorochrome [Gu and Sheppard 1995], (références, exemple de transformation d'un confocal pour (l'imagerie 2-photons): deux photons situés dans le Rouge peuvent exciter un fluorochrome dont la gamme d'onde correspond aux UV. Les dommages opérés généralement par les rayons UV sont donc évités. En outre, cette technique minimise le phénomène d'évanescence (photobleaching) de la sonde et autorise une meilleure pénétration dans le tissu. Enfin, les dommages du laser sont très réduits dans le cas de l'imagerie multi-photons en raison d'une faible émission du signal laser au niveau des plans non focaux.
