There are three different frames(starting a codon with the first, second, or third nucleotide). Generally, only one frame encodes a polypeptide; this frame is specified by the location of the initiation codon of the polypeptide. When one wishes to perform site specific mutagenesis on a gene and re-express the mutant genes into altered proteins, several considerations should be taken initially to increase the efficiency of generating mutants.
