During DNA replication, each of the two strands that make up the double helix serves as a template from which new strands are copied. The new strand will be complementary to the parental or "old" strand. Each new double strand consists of one parental strand and one new daughter strand. In broad outline the plus-and-minus method calls for synthesizing pieces of DNA that are complementary to various regions of the DNA being sequenced. Because of the way the synthesis is manipulated each piece ends either with or just before a particular, identifiable nucleotide(A, G, T or C).
