deconvolution microscope : using the deconvolution microscope, researchers can see the locations of target molecules inside the cell. Seeing where glowing molecules reside within a cell is helpful for a range of research projects--from studies of how the distribution of various proteins changes during an organism's development to investigations of how administration of a drug alters the locations of certain proteins. The microscope, called DeltaVision, is manufactured by Applied Precision, based in Issaquah, Wash. It works by magnifying images and quantifying the intensity of light shed by the sample. The user specifies which colors of light are of interest. For instance, a user can program the microscope to report the intensity of a particular shade of green shining from the cytoplasm of a cell. After a researcher uses the microscope to make a series of optical slices through a specimen, the scope's computer "deconvolves" the images--in other words, it subtracts the out-of-focus light. Then the computer stacks together the images of the slices to reconstruct the original microscope sample. This gives researchers gh-resolution images of the sample's three-dimensional structure.

Confocal microscopy. Unlike "normal" microscopy, only a spot of a minimum size in the specimen is illuminated in confocal microscopy. To enable an image to be formed from this, the specimen must be scanned. The image of the spot is directed through a pinhole stop in an intermediate image plane. As a result, only light from the focal plane can reach the detector(a photomultiplier). All other(out-of-focus) planes are blocked out. This results in an "optical section". The images are stored electronically and displayed on a monitor. A series of optical sections can be recorded by moving a motor a slight distance along the z-axis each time an image has been recorded, after which the next image is then also recorded. Such a z-series permits the electronic reconstruction of the three-dimensional structure using suitable computer programs. The procedure, which is restricted to incident-light techniques, has completely revolutionized fluorescence microscopy in biology in particular.

La localisation de molécules spécifiques, dans le contexte cellulaire, apporte d'importantes contributions à l'étude fonctionnelle des systèmes biologiques à différents niveaux d'organisation : subcellulaire, cellulaire, tissulaire ou organisme entier. L'utilisation de traceurs fluorescents très spécifiques permet de localiser in situ avec une résolution spatiale de l'ordre de 400 - 500 nanomètres, des composants macro-moléculaires tels que acides nucléiques et protéines, mais aussi de petites molécules telles que des peptides, lipides et même des ions.

La microscopie confocale permet la localisation in situ d'une sonde fluorescente dans des cellules d'un spécimen d'une épaisseur de 10 mm à 50 mm selon certains auteurs (5 mm à 30 mm plus raisonnablement), sans sectionnement mécanique préalable. SIAM is a technology that defies the current laws of imaging. It permits scientists to view images much smaller than the wavelength of light and approaching the atomic level, an achievement that has long been considered impossible according to the basic principles of classical optics. SIAM works by shining light through a thin sample onto an ultra-sharp, vibrating tip on the other side. The light scattering from the tip gets altered as the tip approaches within a nanometer from the sample surface, as it vibrates, due to the optical interaction with the local properties of the sample. An image is built up by recording the scattered light as the probe is scanned line by line. The scientists recorded the variations in the scattered light using a very sensitive device--an optical interferometer--which is capable of measuring modulations in light level corresponding to single photons. As the tip moved across the surface, these variations were mapped by a computer into a picture of the surface, showing features such as oil droplets and steps on mica that were only a few atoms in size.

Des forces de liaisons intermoléculaires (appelées forces de Van der Waals) apparaissent alors entre l'atome de matière formant l'extrémité de la micropointe et ceux de l'échantillon. Ces forces modifient les vibrations de la lamelle. Après s'être réfléchi sur l'échantillon et sur la lamelle, le faisceau d'analyse se combine avec le faisceau de référence dans un dispositif interférométrique. Un nouveau faisceau permet d'analyser les modifications des vibrations de la lamelle; il renseigne donc sur le relief de l'échantillon observé. En promenant ce dernier sous la micropointe, on peut reconstruire son image point par point avec une extrême précision.

By orienting and networking such wires, nano-circuitry can potentially be created, which may be of value in the computer industry. The nanostructures can also be incorporated into coatings with optical, electric, magnetic, catalytic, or enzymatic moieties as functional units. A nanoarray is a nanoscale or microscale ordered arrangement of nanotemplates and/or nanostructures. A nanoarray, therefore comprises an ordered array of nanostructures. A nanoarray can have any type of long range packing symmetry, including 2-, 3-, 4-, or 6-fold packing symmetry. The nanoarray can be a one-dimensional structure, a two-dimensional array, or a three-dimensional array. In a specific embodiment, where the nanoparticles are dielectrics, a three-dimensional nanoarray can be a photonic bandgap crystal. Optimal conditions for assembly and crystallization of a nanoarray... are easily determined by routine experimentation, such as by changing each variable individually and monitoring formation of the appropriate products.

We examine how synthetic dynamic nanostructured surfaces control the attachment of pathogenic biofilms. The parameters governing long-range patterning of bacteria on high-aspect-ratio (HAR) nanoarrays are combinatorially elucidated, and we discover that sufficiently low effective stiffness of these HAR arrays mechanoselectively inhibits ~40% of Pseudomonas aeruginosa biofilm attachment. Inspired by the active echinoderm skin, we design and fabricate externally actuated dynamic elastomer surfaces with active surface microtopography.

A confocal laser scanning microscope is a light microscope having a dramatically higher axial resolution than a conventional microscope. In other words, a confocal microscope is capable of optical sectioning. This is accomplished by illuminating the specimen one point at a time with a focused laser beam and using an optical arrangement, where only the light originating from the focus can be detected. The image is formed by scanning the laser beam over the specimen and recording the image into computer memory. A three-dimensional view of the specimen can be reconstructed from a stack of such images taken at successive focal planes.

The Window Observational Research Facility(WORF). The WORF will provide a stable platform such that the full capacity of the U. S. Laboratory window(the highest optical quality window ever flown on an inhabited spacecraft) can be utilized. A conventional animation is created in a fairly fixed sequence : The story for the animation is written(or perhaps merely conceived), then a storyboard is laid out.... Next, the soundtrack(if any) is recorded, a detailed layout is produced(with a drawing for every scene in the animation), and the soundtrack is read-that is, the instants at which significant sounds occur are recorded in order. The detailed layout and the soundtrack are then correlated. Next, certain key frames of the animation are drawn-these are the frames in which the entities being animated are at extreme or characteristic positions, from which their intermediate position can be inferred. The intermediate frames are then filled in(this is called inbetweening), and a trial film is made(a pencil test)... Many stages of conventional animation seem ideally suited to computer assistance, especially inbetweening and coloring, which can be done using the seed-fill techniques.... Before the computer can be used, however, the drawings must be digitized. This can be done by using optical scanning, by tracing the drawings with a data tablet, or by producing the original drawings with a drawing program in the first place. The drawings may need to be postprocessed(e. g., filtered) to clean up any glitches arising from the input process(especially optical scanning), and to smooth the contours somewhat. The composition stage, in which foreground and background figures are combined to generate the individual frames for the final animation, can be done with the image-composition techniques....
