In the carrier-binding mehtod ... care is required regarding the selection of carriers as well as in binding techniques. Namely, the amount of enzyme bound to the carrier and the activity after immobilization depend markedly on the nature of the carrier. The artificial immobilization of enzymes can be subclassified according to the binding mode of the enzyme into :physical adsorption, ionic binding, chelation or metal binding, and covalent binding. Information on changes of enzymatic properties caused by the immobilization of enzymes and microbial cells is useful not only for the application of immobilized systems but also for the elucidation of structure-function relationships and mechanisms of enzyme reaction. The changes of enzymatic properties are considered to be caused by the following two factors. One is changes of the enzyme itself, and the other is physical and chemical properties of the carriers used for immobilization. The physical adsorption method for the immobilization of enzyme is based on the physical adsorption of enzyme protein on the surface of water-insoluble carriers. This method often causes little or no conformational change of the enzyme protein, or destruction of its active center. Accordingly, if a suitable carrier for the target enzyme is found, this method may be both simple and effective.
