A confocal laser scanning microscope is a light microscope having a dramatically higher axial resolution than a conventional microscope. In other words, a confocal microscope is capable of optical sectioning. This is accomplished by illuminating the specimen one point at a time with a focused laser beam and using an optical arrangement, where only the light originating from the focus can be detected. The image is formed by scanning the laser beam over the specimen and recording the image into computer memory. The Binary Colloidal Alloy Test-3 is an Exploration Systems’ transition flight experiment in the Human System Research and Technology area. BCAT-3 provides a unique opportunity to explore fundamental physics and simultaneously develop important future technology, including computers operating on light, complex biomolecular pharmaceuticals, clean sources of geothermal power, and novel rocket engines for interplanetary travel. These studies depend entirely on the microgravity environment provided by the International Space Station(ISS) ;in all other locations accessible to science, gravity dominates and precludes investigation of any other effects of interest. The experiment itself [consists of] photographing samples of colloidal particles with a digital camera onboard the ISS. Colloids are tiny nanoscale spheres of plexiglass a thousand times smaller than the width of a human hair(submicron radius) that are suspended in a fluid. They are ubiquitous(e. g., milk, smoke, and paint) and therefore interesting to study directly. Colloids are also small enough that they behave much like atoms and so can be used to model all sorts of phenomena because their size, shape, and interactions can be controlled. The 10 samples in BCAT-3 are made from the same ingredients, each a recipe with different proportions, and are grouped into three experiments : critical point, binary alloy, and surface crystallization. BCAT-3 is comprised of three separate experiments, which are individually referred to as a binary alloy (BCAT-3-BA), surface crystallization (BCAT-3-SC) and critical point (BCAT-3-CP). The Binary Colloidal Alloy Test-3 (BCAT-3) hardware supported three investigations in which ISS crews photographed samples of colloidal particles (tiny nanoscale spheres suspended in liquid) to document liquid/gas phase changes, growth of binary crystals, and the formation of colloidal crystals confined to a surface. Colloids are small enough that in a microgravity environment without sedimentation and convection, they behave much as atoms and so can be used to model all sorts of phenomena because their size, shape, and interactions can be controlled. The BCAT-3 payload consists of ten small samples of colloid alloys in which the microscopic colloid particles are mixed together into a liquid. These ten samples are contained within a small case that is the size of a school textbook. At the start of an experiment run, all ten samples are shaken to completely remix the colloid samples, much in the same way that salad dressing must be shaken to remix oil and vinegar. After the samples are mixed, what remains is periodically photographed using a digital camera until the colloid and liquid components of those samples have separated or the polymers have formed crystals. The samples can be remixed to repeat the experiment. From a more industrial point of view, whenever multiconstituent liquids are submitted to long-lasting temperature gradients, they are liable to experience thermal diffusive flows, modifying the distribution of the species concentration. Among the cases where this effect has been emphasized, we can mention the elaboration of high technology materials(solidification of semiconductors), the production or purification of light metals... and the optimization of petroleum extraction(by modelling of the hydrocarbon distribution in the reservoirs). So, knowledge of quantitative values of the Soret coefficient(which is the parameter characteristic of the phenomenon) for a wide domain of materials, in various thermodynamic conditions and physical configurations is needed for the above-mentioned basic and applied purposes.... The experimental determination of Soret coefficients in binary mixtures, denoted ST, is obtained by measuring the steady concentration gradient produced by the imposition of a constant and uniform temperature gradient. Le microscope confocal à balayage laser (MCBL) est un microscope optique à éclairage épiscopique géré par un ordinateur qui est capable de sectionner optiquement un échantillon épais. Son image est ensuite reconstruite en superposant toutes ou certaines des sections optiques. Le faisceau lumineux (laser) balaie la surface de l'échantillon au moyen d'un miroir galvanométrique et la lumière réfléchie ou émise (fluorescence) provenant de chaque point de la surface balayée est recueillie par un détecteur, numérisée et stockée dans la mémoire de l'ordinateur. Sur le trajet des rayons provenant de l'échantillon se trouve un diaphragme (pinhole), situé sur l'avant du détecteur dans le plan conjugué avec celui de l'échantillon (confocal). Seuls les rayons image venant du plan focal, qui est confocal avec le plan du diaphragme, parviennent au détecteur. Les autres rayons, provenant d'autres plans focaux, sont bloqués par le diaphragme [...]. L'image résultante obtenue par balayage au niveau du plan focal a une épaisseur extrêmement limitée (section optique) et n'est pas perturbée par des images floues des plans situés au dessus ou au dessous. Comme l'ordinateur déplace la platine dans la direction de l'axe optique Z avec des mouvements micrométriques prédéfinis après chaque balayage, le MCBL ne peut pas recueillir les images provenant d'autres plans. L'ordinateur stocke les images correspondants aux sections optiques successives.
