Muscarinic acetylcholine receptors are members of rhodopsin-like G-protein coupled receptor family. They play several important roles; they mediate many of the effects of acetylcholine in the central and peripheral nervous system and modulate a variety of physiological functions, such as airway, eye and intestinal smooth muscle contraction, heart rate and glandular secretions. The receptors have a widespread tissue distribution and are a major drug target in human disease. They may be effective therapeutic targets in Alzheimer's disease, schizophrenia, Parkinson's disease and chronic obstructive pulmonary disease.

Research is on hairpin catalytic RNA, a catalytic RNA discovered and characterized here in my laboratory at Northern Illinois University. Hairpin catalytic RNA is a molecule which can be engineered to very efficiently cleave target RNA sequences in a unique and efficient reaction. The catalytic RNA can thus be used for the specific down regulation of gene expression since it can be designed to destroy mRNA. Current applications being developed are down regulation of selected genes, including those in the virus HIV-1. Thus it has potential as an AIDS therapeutic.

Nearly every drug molecule works through a structural interaction with a "target" or "receptor" molecule or protein that plays key roles in all biological processes. In the most common model for this interaction, the drug molecule inserts itself into a functionally important crevice of its target protein, like a key in a lock. The molecule then binds there and either induces or, more commonly, inhibits the protein's normal function. This universal drug-target scheme suggests a powerful alternative approach to drug discovery : if it were possible to identify, in advance, the appropriate protein target for a given therapeutic need and if enough were known about the distinguishing structure of that target protein, it ought to be feasible to design the structure of an ideal drug to interact with it.

[Alternative vector systems] include both viral and nonviral approaches to accomplish gene transfer. In both of these approaches, the goal is to develop a system that can deliver genes in vivo after systemic administration. [The] central nervous system agents [belong to] a class of drugs producing both physiological and psychological effects through a variety of mechanisms. They can be divided into "specific" agents, e. g., affecting an identifiable molecular mechanism unique to target cells bearing receptors for that agent, and "nonspecific" agents, those producing effects on different target cells and acting by diverse molecular mechanisms. Those with nonspecific mechanisms are generally further classed according to whether they produce behavioral depression or stimulation. Those with specific mechanisms are classed by locus of action or specific therapeutic use.

Multifunctional nano-devices based on dendritic polymer components will be developed that target neoplastic cells and sense the earliest signatures of cancer. The dendritic nano-devices will be designed to support the specific release of a therapeutic agent within a tumor, and analyze the effect of the therapeutic identifying evidence of residual disease.

C'est pourquoi on pense utiliser des dendrimères pour la libération contrôlée de médicaments. On a constaté aussi que l'ADN s'enroulait autour de certains dendrimères, qui pourraient ainsi servir au transport de gènes en thérapie génique.

Moreover, the administration of genes as therapy requires vehicles--so-called vectors--that encapsulate the gene and guide it to the target cell. The binding of the gene to the cell, its internalization, the transport of the administered genome to the nucleus, and the expression of the gene all constitute potential limitations of this process.

Mais surtout, à côté de la pathologie, un fantastique domaine s'ouvre au transfert et à la thérapie génique, c'est celui du gène considéré comme médicament. La médecine utilise en effet toute une série de molécules d'origine protéique, comme les facteurs antihémophilliques, l'insuline, l'hormone de croissance, pour ne citer que les plus importantes. Le futur pourrait être la fabrication directe par l'organisme des molécules actives après transfert de gènes dans les cellules permissives. Mieux encore, on pourra vraisemblablement dams l'avenir injecter directement par voie intramusculaire par exemple, l'ADN génique, les cellules de l'organisme se comportant ensuite comme de mini-usines de fabrication et de mini-unités de distribution.
