To establish new methods in the lipid research, Far-eastern blot(TLC [thin-layer chromatography] blot), was established for transferring of lipids from an high-performance thin-layer chromatography(HPTLC) plate to a polyvinyledene difluoride(PVDF) membrane within a minute. Applications of this with other methods have been studied. Far-eastern blot, makes possible the following : 1) Purification of glycosphingolipids and phospholipids. 2) Structural analysis of lipids in conjunction with direct mass spectrometry. 3) Binding study using various ligands such as antibodies, lectins, bacterium, viruses, and toxins, and 4) Enzyme reaction on membranes. Not only analysis of lipids but also metabolites of drugs and natural compounds from plants, and environmental hormones are possible by this method.

Le principe de ces détecteurs est le suivant : à la sortie de la colonne de chromatographie on adapte un diviseur d'effluents ayant un rapport de division de 90/10 par exemple; 10 % des effluents sont envoyés au détecteur afin d'enregistrer leur sortie et de les repérer : 90 % des effluents sont envoyés par un tube capillaire chauffé dans une chambre à combustion. Seul le CO2 marqué parvient à une chambre de comptage proportionnel. Cette chambre est composée d'une enceinte parfaitement propre traversée en son centre par un filament de tungstène porté à une tension de 1 500 volts en courant continu. Les ions libérés sous l'effet de la radioactivité sont collectés sur la paroi interne de la chambre de comptage et le signal ainsi obtenu est envoyé après intégration à un enregistreur.

The detection of spots is sometimes easier than with paper chromatography because more universal techniques can be used. Frequently, colorless or nonfluorescent spots can be visualized by exposing the developed plate to iodine vapor. The iodine vapor interacts with the sample components, either chemically or by solubility, to produce a color. Thin-layer plates and sheets are commercially available which incorporate a fluorescent dye in the powdered adsorbent. When held under ultraviolet light, dark spots appear where sample spots occur due to quenching of the plate fluorescence.
