Immobilization of enzymes by crystallization and subsequent cross-linking provides structures which have a regular three-dimensional molecular arrangement, and a high packing density. Such structures are useful for both X-ray diffraction analysis, and also physico-chemical studies of enzymes in the solid state. The analytical electron microscope(AEM) is based on a conventional 200 keV transmission electron microscope(TEM) which has been augmented to include the functions of a scanning transmission electron microscope(STEM) and the scanning electron microscope(SEM). Since the electrons must penetrate through the specimen, the specimen dimensions must be no greater than 1 micrometer thick, and preferably in the range of 100 nm. Bulk specimens may be thinned to this range by a combination of mechanical, chemical, and ion sputtering techniques, all of which are available in the microanalysis laboratory. As an imaging tool, the AEM is capable of 0. 3 nm spatial resolution in the TEM mode, 1 nm resolution in the STEM mode, and 3 nm resolution in the SEM mode. Elemental microanalysis is carried out by means of energy dispersive x-ray spectrometry(EDS) and electron energy loss spectrometry(EELS), which together provide sensitivity to all elements with atomic number > 2. X-ray powder diffraction is used in the identification of crystalline phases in unknown samples, finding the lattice parameter and other lattice related information, quantitative determination of phases in multiphase samples, finding the orientation in single crystals, and stereographic projections. The samples must be crystalline solids such as metals, ceramics, rocks etc. These samples are usually about 1 mg in size. If the sample fits in the sample holder then there is no preparation necessary otherwise the sample may have to be crushed in order to fit. The sample takes less than one hour to be analyzed for qualitative analysis in major phases and up to 16 hours for trace phases. The quantitative analysis can take several minutes to several hours after set up. Powder X-Ray Diffraction. Powders of crystalline materials diffract X-Rays. A beam of X-rays passing through a sample of randomly-oriented micro-crystals produces a pattern of rings on a distant screen. Powder X-Ray diffraction provides less information than single-crystal diffraction, however, it is much simpler and faster. Powder X-Ray diffraction is useful for confirming the identity of a solid material and determining crystallinity and phase purity. Modern powder X-Ray diffractometers consist of an X-Ray source, a movable sample platform, an X-Ray detector, and associated computer-controlled electronics. X-ray powder diffraction analysis(XRPD) was proven to be a very powerful tool for the study of "green stone" Neolithic axes and other artifacts. The amount of material to be sampled can be reduced to a few milligrams, thus making it a quasi-non destructive method.... If the dimension of an object is adequate(up to 60 cm in bulk, depending upon the shape) it can be directly analyzed, by XRPD, without sampling. Even a rough, irregular surface, both on flat and bent objects, is suitable for the analysis. In the last 10-15 years many analytical advances in X-ray fluorescence analysis(XRF) have taken place, giving rise to non-destructive ultrasensitive surface analyses of materials. One of the variants of XRF developed is micro-XRF(5-XRF), which is able to analyse the distribution of major, minor and trace elements in microscopic sample surface areas. La micro-fluorescence X est une technique qui consiste à exciter par un micro-faisceau de rayons X les seuils d'absorption des éléments chimiques présents dans un échantillon. La désexcitation de ces atomes induit l'émission de photons dont l'énergie est caractéristique de l'élément en question. Ces photons sont collectés par un détecteur et classés dans des canaux en fonction de leur énergie. On obtient ainsi ce qu'on appelle un spectre d'émission de fluorescence. Ce spectre comporte des pics (ou raies) qui caractérisent les différents éléments présents dans l'échantillon. Tout l'échantillon est balayé point par point par le micro-faisceau et un spectre d'émission est acquis pour chaque point.
