To solve the problem on the microstructural characterization of metallic superlattices, taking the NiFe/Cu superlattices as example, we show that the structures of metallic superlattices can be characterized exactly by combining low-angle X-ray diffraction with high-angle X-ray diffraction. First, we determine exactly the total film thickness by a straightforward and precise method based on a modified Bragg law from the subsidiary maxima around the low-angle X-ray diffraction peak. Then, by combining with the simulation of high-angle X-ray diffraction, we obtain the structural parameters such as the superlattice period, the sublayer and buffer thickness. Immobilization of enzymes by crystallization and subsequent cross-linking provides structures which have a regular three-dimensional molecular arrangement, and a high packing density. Such structures are useful for both X-ray diffraction analysis, and also physico-chemical studies of enzymes in the solid state. An important feature of the enzyme crystals is that they have large liquid-filled channels surrounding the individual protein molecules, therefore, as in dilute solution, most of the surface of the enzyme is in contact with the surrounding solvent. Since the channels are permeable to low molecular weight solutes, it is possible to study the properties of enzymes in crystals in much the same way as when in solution. Lysosyme is an... enzyme whose catalytic mechanism is being studied. Groutite occurs in two distinct habits. The first of these, reported by Klein and Frondel(1967), occurs as platy, irregular, bladed, opaque 0. 5 mm crystals, black, with strong reddish-brown internal reflections and perfect {010} cleavage(Figure 22-57). X-ray powder diffraction is used in the identification of crystalline phases in unknown samples, finding the lattice parameter and other lattice related information, quantitative determination of phases in multiphase samples, finding the orientation in single crystals, and stereographic projections. The samples must be crystalline solids such as metals, ceramics, rocks etc. These samples are usually about 1 mg in size. If the sample fits in the sample holder then there is no preparation necessary otherwise the sample may have to be crushed in order to fit. The sample takes less than one hour to be analyzed for qualitative analysis in major phases and up to 16 hours for trace phases. Powder X-Ray Diffraction. Powders of crystalline materials diffract X-Rays. A beam of X-rays passing through a sample of randomly-oriented micro-crystals produces a pattern of rings on a distant screen. The X-ray diffraction results, in fact, point to a new form of structural disorder called phason disorder, which is unique to quasicrystals. If one compares a Penrose tiling or a quasiperiodic sequence with a conventional crystal, one sees that the quasiperiodic structures have the ability to generate a new form of disorder during growth : a defect can arise when the wrong kind of unit cell, or line segment, falls in a particular place. In the technique of X-ray diffraction the X-rays are allowed to strike a protein crystal; the X-rays, diffracted(bent) by the crystal, impinge on a photographic plate, forming a pattern of spots. This method reveals that peptide chains can assume very complicated, apparently irregular shapes. A good indication of the positions of nearly all the non-hydrogen atoms in a protein can be obtained from a structure solved to a resolution of 3. 0 A or better. Such a study involves a series of stages. Firstly, tens of milligrams of the protein must be isolated in pure form, and then crystals of millimeter dimensions must be grown which diffract X-rays to high resolution. However, reconstruction of the electron density of the molecule from diffraction data requires information about the amplitude and the phase of the X-ray reflections. To solve the phase problem, diffraction data from at least one heavy-metal derivative of the protein must be measured.
