High brightness, short pulse width(<200 picoseconds) x-ray lasers now offer biologists the possibility of high-resolution imaging of specimens in an aqueous environment without the blurring effects associated with natural motion or chemical erosion. As a step toward developing the capabilities of this type of x-ray microscopy, a tantalum x-ray laser at a 44. 83-angstrom wavelength was used together with an x-ray zone plate optic to image both unlabeled and selectively gold-labeled dried rat sperm nuclei. In the image, 500-angstrom features are clearly visible. This illustrates the importance of x-ray microscopy in determining chemical composition. X-ray microscopy also provides information about the uniformity of sperm chromatin organization and the extent of sperm chromatin hydration. 2, record 2, English, - X%2Dray%20microscopy

The popularity of cryoelectron microscopy stems from the fact that it allows the observation of specimens that have not been stained or fixed in any way, showing them in their native environment, in contrast to X-ray crystallography, which generally requires placing the samples in non-physiological environments, which can occasionally lead to functionally irrelevant conformational changes. 2, record 3, English, - cryo%2Delectron%20microscopy

2, record 5, French, - microscopie%20%C3%A9lectronique

Amyloidosis is not one clinical entity but a group of diverse structurally driven protein deposition diseases. They are similar in that protein deposition occurs extracellularly and these deposits stain eosinophilic using standard tissue histologic stains, bind Congo red dye, and emit an apple-green birefringence using polarized light microscopy; exhibit metachromasia with crystal violet; and have an array of 75-to 100-Å nonbranching fibrils by electron microscopy and a twisted b-pleated sheet antiparallel configuration by x-ray crystallography. They differ, however, in the biochemical nature of the proteinaceous deposits, the "etiology" of the associated diseases(neoplastic, inflammatory, degenerative, hereditary), the tropism of protein deposition, and the spectrum of disease manifestations. 1, record 6, English, - amyloidosis

Every element absorbs X rays in a distinctive manner. At X-ray wavelengths between 23 and 44 angstroms, oxygen-and therefore water-is much more transparent to radiation than is organic material. This fact opens up the possibility of discerning features through water, which makes up about three quarters of the mass of most cells. This part of the X-ray spectrum, called the water window, is quite useful for biological microscopy. 1, record 11, English, - water%20window

... the first images were made by shining X rays through a specimen placed in contact with ordinary photographic film. The X rays that passed through the specimen exposed the film, and the resulting image could then be examined with a light microscope. 1, record 12, Spanish, - microradiograf%C3%ADa%20de%20contacto
