A more detailed map of a large YAC [yeast artificial chromosome] insert can be produced by subcloning, a process in which fragments of the original insert are cloned into smaller-insert vectors. Because some YAC regions are unstable, large-capacity bacterial vectors(i. e. those that accommodate large inserts) are also being developed. 1, record 1, English, - large%2Dcapacity%20bacterial%20vector

We therefore propose to undertake the identification, assembly and test of the structural elements necessary for the proper function and stability of a mammalian artificial chromosome(MAC). To achieve this goal, we will create a chimaera between an existing Yeast Artificial Chromosome(YAC) and selected human and other mammalian genomic sequences. The stability and autonomous replication of the MAC in human cells will be evaluated by a variety of assays, both before and after the addition of candidate origins, telomeres and centromeres. 2, record 2, English, - mammalian%20artificial%20chromosome

3, record 2, French, - chromosome%20mammif%C3%A8re%20artificiel

Individual primary recombinant clones(hosted in phage, cosmid, YAC, or other vector) that are placed in two-dimensional arrays in microtiter dishes. Each primary clone can be identified by the identity of the plate and the clone location(row and column) on that plate. Arrayed libraries of clones can be used for many applications, including screening for a specific gene or genomic region of interest as well as for physical. 1, record 3, English, - arrayed%20library

4, record 4, French, - chromosome%20artificiel%20de%20levure

Clusters of genes with one per 15 or 30 kilobases were described, but they also noted that only a single cDNA was captured from YAC clones spanning a 6 megabase region of the q21 band by selection methods. Why so few genes were present in such a large region, which corresponded to a darkly staining Giemsa band, appeared to be an intriguing question and one that remained unresolved. 1, record 5, English, - Giemsa%20band

1, record 5, French, - bande%20Giemsa
