Deoxyribonuclease(DNAse) agar actually has DNA in it. Deoxyribonuclease is an exoenzyme excreted from the cell which will break the DNA down into smaller molecules. There are 2 kinds of DNAse agar that can be used. 1) The first medium has no indicator, but is does require HCl(hydrochloric acid) reagent to be added after growth has occurred on the plate. Without DNAse enzyme, the HCl precipitates out the undestroyed DNA. The rest of the agar with the remaining DNA, which binds to HCl will have turned cloudy. Around the growth area, if the bacterium has made DNAse and destroyed the DNA, there will be clear zones seen. 2) DNAse agar with the indicator methyl green can be used. In this medium, when the DNA in the medium is hydrolyzed by the bacterial DNAse enzyme, the pieces of cut DNA release from binding to methyl green, and you will see a yellowish/orange zone around the growth area. Lock-in is a fundamental physical phenomenon associated with all oscillating devices. Laser gyros exibit the same phenomenon, in that the coupling mechanism is the reflection of light from one laser beam back along the path where it interacts with the oppositley traveling beam. This is called backscatter and is caused primarily by imperfect mirrors. Therefore, at low rotation rates, there is no frequency difference between the counter-rotating beams so that the gyroscope does not indicate any rotation. Drip Irrigation.... Water is delivered in small quantities under low pressure directly to... the root zone of the plants. You’ll need 1/2 inch polyethylene header pipe, and a roll of thin tape-like porous tubing. The header pipe connects to a garden hose and routes water through the garden. The pipe is placed either down the center or along one end of the garden. Small puncture holes are then made with a screwdriver into the header pipe at intervals corresponding to garden row. Eight-to ten-inch pieces of rubber connecting hose are then inserted into these holes. The other end of the connecting hose attaches to a length of porous tape. This tape is rolled out along each vegetable bed and cut to the same length as the row. Then end of the tape is then crimped to prevent water loss when the system is in use. Reactors. The most widely applied reactors for gasification of biomass are the moving bed types and the fluidized bed types.... Moving bed-counter current. The moving bed reactor is a vertical shaft through which the biomass flows slowly downwards as a packed bed. The reactor has a gastight feeding system at the top and an ash removal system at the bottom. The moving bed reactor is used for biomass having dimensions of 1 to 10 cm. Three sub-types can be distinguished : cocurrent, countercurrent and cross-current reactors. The countercurrent moving bed gasifier is the most simple reactor type. The gas, produced in the reduction zone, leaves the reactor together with the pyrolysis products and the steam from the drying zone. The producer gas is rich in hydrocarbons and it has a temperature of about 400°C. The gas can be used for direct heating purposes.
