Afin de déchiffrer les nombreuses mutations affectant la région régulatrice de l'opéron lactose, W. Gilbert a senti la nécessité de développer une technique de séquence plus rapide que celle de Sanger. Avec son collaborateur, A Maxam, ils ont réussi à trouver des réactions chimiques interrompant spécifiquement la chaîne de DNA en position A, T, G ou C. Ainsi le DNA, marqué à son extrémité 5' par le 32P, est dégradé partiellement et spécifiquement au cours de quatre réactions séparées.
