Dr. Robson's laboratory is studying some of the important house-keeping genes of P. falciparum which cause severe malaria in man. Binding of calcium to calmodulin causes a conformational change, converting it to an active form that can bind to various effector enzymes, converting them in turn to an active conformation. Both cyclases are activated by the host calmodulin. Their genes have been cloned and expressed in E. coli... using an original technique, using E. coli cells expressing one of the components of the activation system for the screening of a library of the pathogen's genome. Once in the cell, calcium binds to a protein called calmodulin, which amplifies the activation of the enzyme adenylyl cyclase by serotonin. When calcium/calmodulin binds to the adenylyl cyclase, the enzyme generates more cyclic AMP. This capacity makes adenylyl cyclase an important convergence site for the conditioned and the unconditioned stimuli.
