There are several types of electrophoresis, zone electrophoresis being one of the most common. In zone electrophoresis, proteins are supported on a solid so that, in addition to the electric migration forces, conventional chromatographic forces may enter into the separation efficiency. There are several types of zone electrophoresis according to the different supports. The common supports include starch gels, polyacrylamide gels, polyurethane foam, and paper.

Chromatography based on the specific chemical(covalent) reaction of the chromatographic material with only one of the components of a mixture. After removal of the other components by washing, the covalently bonded component is released by reaction with a suitable reagent. The reaction that releases the component of interest should preferably leave the chromatographic material in a form that can easily be reactivated. This type of chromatography differs from conventional affinity chromatography by its reliance on scission and formation of covalent bonds during the separation process and by the lack of a requirement for specific adsorptive binding areas of the activating ligand.

Used mainly in the study of proteins and peptides although it has been used also in the study of mercurated polynucleotides. It provides considerable economies in both time and materials.

Chromatographie basée sur la formation d'une liaison chimique. Elle met à profit une réaction largement utilisée dans la nature pour la réticulation des chaînes peptidiques. Il s'agit de la condensation de deux thiols (les cystéines en synthèse de protéines) pour former un pont disulfure R-S-S-R. La liaison entre les deux atomes de soufre est facilement réductible. Silica Gel Column Chromatography.... Getting reproducibility when doing silica gel(or alumina) separations with column chromatography can be a problem. If you do not carefully control the water/moisture content of the silica gel and the mobile phase, moisture is added to or removed from the silica gel during the separation. This results in retention times that either increase(with water addition to the silica gel) or decrease(with water loss from the silica gel).... The reseacher attempting to optimize the flash chromatography of new compounds should investigate the effect of moisture on the separation being attempted. Various glass chromatographic columns containing silica gel with different amounts of moisture can be run side by side to see which column produces the optimum results.

In gas chromatography, the theory that the column operated similarly to a distillation column; for example, chromatographic columns are considered as consisting of a number of theoretical plates, each performing a partial separation of components.

The plate concept of chromatography is inherited from distillation theory, where originally the more efficient distillation columns actually contained discrete plates. Martin and Synge considered a chromatographic system to be composed of a number of imaginary or theoretical plates. However, the number of theoretical plates that a column appears to contain is a poor measure of chromatographic performance since the number of plates is independent of the system dimensions.
