Liquid gel chromatography(LGC) is an established laboratory technique for the differential fractionation and separation of molecules. The underlying premise for simple LGC is that molecules elute from a gel bed in order of decreasing molecular weight. Generally, gel particles fill a chromatography column to form a bed. Type of chromatography developed following the observation that certain proteins were unexpectedly retained on affinity gels containing hydrophobic spacer arms. This concept was extended, and families of hydrophobic adsorbents prepared using a homologous series of hydrocarbon chains of different lengths. In practice most proteins can be satisfactorily purified using C-8 or phenyl substituents. Hydrophobic interactions are strongest at high ionic strength, so adsorption to a hydrophobic matrix can be conveniently performed after a salt precipitation or ion exchange chromatography step. When carrying out the hydrophobic chromatography of human urokinase..., the solution containing the crude urokinase which is prepared from human urine in the usual simple method, is adjusted to a salt concentration of 0. 2 to 2 M, preferably 0. 2 to 1 M. The solution is then made to flow through the hydrophobic column packed with the protein adsorbent of this process.... In 1956, Harley and Pretorius described a simple discharge detector for gas chromatography based on the electrical properties of a glow discharge, the voltage across the gaseous discharge being a function of the type of gas present. Druyvesteyn and Penning in 1940 reviewed the complex nature of the glow discharge, noting that the discharge is maintained by electrons emitted from the cathode which result from processes related to positive ion bombardment, photoelectric emission, and collisions with excited atoms. The most universal continuous detector for liquid chromatography is the differential refractometer. This detector is universal only to the extent that one can select an eluent the refractive index of which differs from that of the solutes. Pure eluent flows through one cell of the detector while column effluent flows through the other cell. When a solute with a refractive index different from that of the solvent enters the detector, an imbalance occurs, which is transmitted to the recorder. Two strategies have been used to clone specific sequences of eukaryotic, genomic DNA in bacteriophage lambda vectors. The first involved cloning of DNA preparations that had been highly enriched for the sequences of interest. Total genomic DNA was digested to completion with a restriction endonuclease and the resulting fragments were separated by preparative gel electrophoresis, reverse-phase column chromatography, or both. The fractions containing the DNA sequences of interest were identified by hybridization to appropriate probes and cloned in bacteriophage lambda vectors. Because these fractionation procedures may yield an overall enrichment of as much as 100-fold to 300-fold, the number of recombinant bacteriophages that had to be generated and screened for the DNA sequences of interest could be significantly reduced.
