A partition process in which the paper acts both as the support and, by virtue of the water held in its fibres, as the stationary phase.

The tendency for a compound to divide its time between two immiscible solvents(solvents such as hexane and water which won’t mix) is known as partition. Paper chromatography using a non-polar solvent is therefore a type of partition chromatography.

Paper chromatography... is used widely for qualitative identification, although quantitative analysis can be done. A sample is spotted onto a strip of filter paper with a micropipet, and the chromatogram is "developed" by placing the bottom of the paper(but not the sample spot) in a suitable solvent... The solvent is drawn up the paper by capillary action, and the sample components move up the paper at different rates, depending on their solubility and their degree of retention by the paper.

[The method] was innovative in that it involved the concept of a stationary liquid (water) supported on an inert solid (silica), with the result that the solute molecules partitioned between the stationary liquid and a separate mobile liquid phase (chloroform). The technique came to be called partition chromatography.

La chromatographie de partage [...] a d'abord été utilisée pour la séparation des aminoacides [...] et a été depuis appliquée à des problèmes de séparation de différentes substances. La séparation est effectuée dans un nombre immense d'étapes de partage isolées, chacune de dimension microscopique dans une colonne (10 à 100 cm de longueur) remplie de grains d'une substance hydratée, insoluble et inerte, comme l'amidon ou le gel de silice. Les grains d'amidon ou de gel de silice sont hydrophiles et sont entourés par une phase d'eau finement liée, qui sert de phase stationnaire aqueuse à travers laquelle passe un flux de solvant non miscible qui traverse la colonne par gravité. Chaque grain d'amidon agit ainsi comme un séparateur microscopique.

A physical method of separation in which the components to be separated are distributed between two phases, one of which is stationary (stationary phase) while the other (the mobile phase) moves in a definite direction.

OBS

There are two basic types of chromatography, gas chromatography and liquid chromatography. Gas chromatography can be subdivided into two categories : gas-solid chromatography and gas-liquid chromatography. Similarly, liquid chromatography comprises column chromatography and planar chromatography. In turn, these two classes are broken up as follows : planar chromatography with two sub-classes : thin layer chromatography and paper chromatography. As for column chromatography it contains five sub-classes : sub-class 1 :(liquid-solid chromatography, flash chromatography, supercritical fluid chromatography and high-pressure liquid chromatography), sub-class 2(liquid-liquid chromatography, droplet counter current chromatography), sub-class 3(bonded phase chromatography), sub-class 4(ion exchange chromatography), sub-class 5(exclusion chromatography which includes gel permeation chromatography and gel filtration chromatography).

A zone in paper and thin-layer chromatography of approximately circular appearance.

The detection of spots is sometimes easier than with paper chromatography because more universal techniques can be used. Frequently, colorless or nonfluorescent spots can be visualized by exposing the developed plate to iodine vapor. The iodine vapor interacts with the sample components, either chemically or by solubility, to produce a color. Thin-layer plates and sheets are commercially available which incorporate a fluorescent dye in the powdered adsorbent. When held under ultraviolet light, dark spots appear where sample spots occur due to quenching of the plate fluorescence.
