To establish new methods in the lipid research, Far-eastern blot(TLC [thin-layer chromatography] blot), was established for transferring of lipids from an high-performance thin-layer chromatography(HPTLC) plate to a polyvinyledene difluoride(PVDF) membrane within a minute. Applications of this with other methods have been studied. Far-eastern blot, makes possible the following : 1) Purification of glycosphingolipids and phospholipids. 2) Structural analysis of lipids in conjunction with direct mass spectrometry. 3) Binding study using various ligands such as antibodies, lectins, bacterium, viruses, and toxins, and 4) Enzyme reaction on membranes. Not only analysis of lipids but also metabolites of drugs and natural compounds from plants, and environmental hormones are possible by this method.

Cette carte bidimensionnelle est obtenue en effectuant une électrophorèse d'un mélange peptidique selon une direction de la feuille de papier, en séchant le papier, puis en effectuant une séparation chromatographique des peptides dans l'autre direction. Chromatography in which the stationary phase is a thin layer of adsorbent gel coated on a glass or plastic plate. The sample is applied in a small spot at the base of the plate, and the plate is made to stand on edge in a solvent. As the solvent rises in the gel by capillary action, the sample components move at different rates and are separated into different spots. The plate is removed from the solvent, dried, and stained to visualize the components.

A zone in paper and thin-layer chromatography of approximately circular appearance.

The detection of spots is sometimes easier than with paper chromatography because more universal techniques can be used. Frequently, colorless or nonfluorescent spots can be visualized by exposing the developed plate to iodine vapor. The iodine vapor interacts with the sample components, either chemically or by solubility, to produce a color. Thin-layer plates and sheets are commercially available which incorporate a fluorescent dye in the powdered adsorbent. When held under ultraviolet light, dark spots appear where sample spots occur due to quenching of the plate fluorescence.

In gas chromatography, the theory that the column operated similarly to a distillation column; for example, chromatographic columns are considered as consisting of a number of theoretical plates, each performing a partial separation of components.

The plate concept of chromatography is inherited from distillation theory, where originally the more efficient distillation columns actually contained discrete plates. Martin and Synge considered a chromatographic system to be composed of a number of imaginary or theoretical plates. However, the number of theoretical plates that a column appears to contain is a poor measure of chromatographic performance since the number of plates is independent of the system dimensions.

While the plate theory and the various rate theories of chromatography describe the distribution of a solute in the system, selectivity is a consideration of the relative distributions of two or more solutes in a chromatographic process. The ability of chromatography to separate two solutes depends on the selectivity of the process and the degree to which the system can distinguish thermodynamically between the solutes. Selectivity, then, is simply a measure of the relative retention or relative mobilities of two solutes.
