Eddy diffusion broadens the band when passing through the column and detected in the chromatography. This is due to the molecules of the sample traveling through the column along many different paths of different lengths. The shortest path is the front of the band and the longest path is located in the back of the band. This can be corrected if the band is broad and the column support is of small size and uniform. This effect may be described by the van Deemter equation. H=A+(B/u) +Cu. Where M is the flow rate, A is the eddy diffusion term, B/u is the longitudinal diffusion term and Cu is the mass-transfer rate term. An injection port is necessary in all g. c. [gas chromatography] and h. p. l. c. [high-performance liquid chromatography] systems in which syringe injection is employed. The injection port joins the top of the column to the mobile phase supply line, and has a third outlet through a small orifice covered by a self-sealing septum. Injection of samples is done by inserting the hypodermic syringe containing the sample through this septum so that the point of the needle is immediately above the head of the column. The detection of spots is sometimes easier than with paper chromatography because more universal techniques can be used. Frequently, colorless or nonfluorescent spots can be visualized by exposing the developed plate to iodine vapor. The iodine vapor interacts with the sample components, either chemically or by solubility, to produce a color. Thin-layer plates and sheets are commercially available which incorporate a fluorescent dye in the powdered adsorbent.
