A tissue culture medium containing hypoxanthine, aminopterin, and thymidine, used in somatic cell fusion experiments. Aminopterin(an antifolate) blocks de novo synthesis of purine and thymine nucleotides, but these compounds can be produced from hypoxanthine and thymidine by normal cells possessing the enzymes hypoxanthine phosphoribosyltransferase(HPRT) and thymidine kinase(TK).

Milieu sélectif contenant de l'hypoxanthine, de l'aminoptérine et de la thymidine. La synthèse de l'ADN à partir des sucres et des acides aminés est bloquée par l'aminoptérine. Une autre voie de biosynthèse possible utilise des nucléotides préformés (les précurseurs de l'ADN) et deux enzymes : la thymidine-kinase (TK) et l'hypoxanthine guanine phosphoriboxyl transférase (HGPRT). Les cellules qui possèdent ces enzymes peuvent croître sur un milieu contenant l'aminoptérine et les nucléosides.

Here we describe changes in dNTP metabolism that precede DNA fragmentation in a model of apoptosis driven by deprivation of the cytokine interleukin 3(IL-3). In haemopoietic BAF3 cells, IL-3 withdrawal leads to a rapid decrease in the size of dATP, dTTP and dGTP pools without affecting dCTP levels. This imbalance in dNTP pools precedes DNA fragmentation and is accompanied by down-regulation of enzymes controlling the de novo and salvage pathways of dNTP synthesis, ribonucleotide reductase and thymidine kinase(TK) respectively.

Fatty Acid Synthesis. The cytoplasmic system catalyzing the de novo synthesis of fatty acid is a complex containing all the enzymes required for the conversion of acetyl CoA and malonyl CoA to palmitic acid.... Malonyl CoA is the first product synthesized in the sequence of reactions leading to long-chain fatty acid formation. The reaction is catalyzed by acetyl CoA carboxylase.
