OBS

Denaturation involves the breaking of many of the weak linkages, or bonds(e. g. hydrogen bonds), within a protein molecule that are responsible for the highly ordered structure of the protein in its natural(native) state. Denatured proteins have a looser, more random structure; most are insoluble.

The aggregation of protein molecules into long fibrils is a property of many proteins when they become damaged or denatured. Such aggregates are formed as part of the pathology of many diseases, including Alzheimer's Disease and Parkinson's Disease. It is very important therefore to understand the fundamental reasons why proteins form such structures. Such understanding may eventually lead to therapies based upon interfering with the growth of the protein fibres.

A technique used to identify and isolate the protein product of a cloned gene.

OBS

The cloned double-stranded DNA is denatured and annealed to a mixture of mRNAs known to contain mRNA [messenger RNA] : DNA hybrids are formed, which renders the relevant mRNA unavailable for translation. Translation of the annealed mixture in an in vitro translation system thus results in the absence of translation product of the test gene. Comparison with the translation products of an untreated sample can then identify the missing protein.

In meiotic cells, a protein with broad phyletic distribution and unique cellular localization which shows a high binding efficiency for single-stranded DNA and the property of catalyzing the renaturation of thermally denatured DNA. The protein is assumed to play a role in meiotic recombination and to promote chromosome pairing and crossing over between homologous chromosomes. It is absent from somatic tissues.

La régulation de l'expression d'un gène passe, entre autres mécanismes, par l'association de protéines à certaines des séquences d'acides nucléiques qui le constituent. L'analyse de ces séquences et celles des protéines régulatrices qui s'y fixent est donc d'une importance capitale pour la compréhension des mécanismes de régulation de l'expression des gènes.

One current proteomic tool for visualizing and quantitating all proteins expressed in a ological system at a given time is two-dimensional gel electrophoresis(2-DGE). As originally described by Patrick O’Farrell for analyzing Escherichia coli proteins in 1975, 2-DGE combines the electrophoretic separation of denatured proteins bysoelectric point charge differences in the first dimension with separation based on molecular size differences in the second dimension. The proteins, which can be detected using protein-specific stains, appear as constellations of spots in the 2-D space of the gel. Over the 20-year history of 2-DGE, numerous algorithms have been developed for comparing 2-DGE patterns and quantitatively analyzing protein abundance.
